免疫炎癥反應(yīng)和內(nèi)皮祖細胞在肉雞肺血管叢樣病變形成中的作用(英文)

目的:肺血管叢樣病變是重度肺動脈高壓(PAH)病人的特征病變,病變血管周圍常伴有單個核細胞浸潤。肺血管叢樣病變在常用實驗動物上難以復(fù)制,但可在肉雞(一種生長快速的肉用型雞)肺臟中自發(fā)形成。內(nèi)皮祖細胞(EPCs)在組織再生和血管修復(fù)過程中發(fā)揮重要作用。本研究以肉雞為模型,探討EPCs與肺血管叢樣病變形成之間的關(guān)系。創(chuàng)新點:證實EPCs參與了肺血管叢樣病變的形成過程,并揭示了導致EPCs功能障礙的免疫學機制。方法:采集14周齡肉雞肺組織,常規(guī)石蠟切片,觀察肺血管叢樣病變的形成情況;采用免疫組化法檢測EPCs表面標志CD133和VEGFR-2的表達以及肝細胞生長因子(HGF)的表達;分離培養(yǎng)晚期EPCs,建立EPCs/淋巴細胞共培養(yǎng)體系,并在共培養(yǎng)體系中添加20 ng/ml HGF,觀察EPCs增殖、凋亡和體外管樣結(jié)構(gòu)形成的變化。結(jié)論:在不同周齡的肉雞肺組織中均可觀察到處于不同發(fā)展階段的肺血管叢樣病變(圖1)。早期的叢樣病變主要由EPCs(CD133+和VEGFR-2+細胞)構(gòu)成,HGF在病變實體中高表達(圖2)。淋巴細胞共培...
