【摘要】：為研究禽呼腸孤病毒(ARV)在雞肝癌細(xì)胞(LMH)中的增殖特性,本研究將ARV S1133毒株感染LMH細(xì)胞,連續(xù)傳代3次,觀察細(xì)胞病變(CPE),通過RT-PCR鑒定病毒,并進(jìn)一步研究病毒的增殖特性。結(jié)果顯示,ARV S1133毒株能夠在LMH細(xì)胞中很好地增殖,并出現(xiàn)典型的CPE。一步生長曲線結(jié)果顯示,S1133感染LMH細(xì)胞后的前12 h為潛伏期,12 h~24 h為快速增長期,第24小時的效價達(dá)到最大,TCID50為10-9.2/0.1 m L。本研究為進(jìn)一步研究ARV的致病機(jī)理和研制細(xì)胞滅活疫苗提供了良好的操作平臺。
[Abstract]:In order to study the proliferation characteristics of avian reovirus (ARV) in chicken liver cancer cell line (LMH), LMH cells were infected with ARV S1133 strain for three consecutive generations. The cytopathic (CPE), was observed to identify the virus by RT-PCR, and the proliferation characteristics of the virus were further studied. The results showed that ARV S1133 strain could proliferate well in LMH cells and appeared typical CPE.. The results of one-step growth curve showed that the incubation period of LMH cells was 12 hours after S1133 infection, and the rapid growth period was 24 hours after S1133 infection. The titer of S1133 cells reached the maximum at 24 hours, and the titer of S1133 cells was 10 鈮，
