恩拉霉素高效液相色譜(HPLC)檢測(cè)方法的建立

【摘要】:恩拉霉素是由放線菌(Streptomyces fungicidicus)發(fā)酵而得的一類多肽類抗生素,由于其獨(dú)特的分子結(jié)構(gòu),對(duì)革蘭氏陽(yáng)性菌有很強(qiáng)的活性,特別是對(duì)腸道內(nèi)的有害梭菌有很強(qiáng)的抑殺作用。作為全新的飼料添加劑,恩拉霉素能促進(jìn)豬、雞等的生長(zhǎng),廣泛用于畜牧業(yè)。目前恩拉霉素的含量測(cè)定是按照農(nóng)業(yè)部1999年頒發(fā)的《進(jìn)口獸藥質(zhì)量標(biāo)準(zhǔn)》方法進(jìn)行,本研究根據(jù)恩拉霉素的結(jié)構(gòu)特點(diǎn),通過(guò)實(shí)驗(yàn),建立了恩拉霉素的高效液相色譜檢測(cè)法。篩選優(yōu)化色譜條件為:色譜柱:Thermo C18,5μm,4.6mm×250mm;流動(dòng)相:乙腈:0.05mol/L磷酸二氫鈉溶液=30:70;紫外檢測(cè)波長(zhǎng)267nm;柱溫:30℃;流速:1.0mL/min;進(jìn)樣量:20μL。在該條件下,檢測(cè)結(jié)果顯示恩拉霉素A和恩拉霉素B在12.5~200μg/mL范圍內(nèi)線性關(guān)系良好,恩拉霉素A和恩拉霉素B的回歸方程分別為A1=11587x+18926(R2=0.998)和A2=35969x-46745(R2=0.996);恩拉霉素A的平均回收率為98.9%,恩拉霉素B的平均回收率為98.7%;相對(duì)標(biāo)準(zhǔn)偏差(RSD)為0.27%。實(shí)驗(yàn)結(jié)果證明采用建立的高效液相色譜法(HPLC)檢測(cè)恩拉霉素含量具有線性范圍寬、分析時(shí)間短、樣品前處理簡(jiǎn)便、定量結(jié)果準(zhǔn)確、重復(fù)性好、穩(wěn)定性好和回收率高等特點(diǎn),可以作為恩拉霉素含量檢測(cè)的質(zhì)控方法。同時(shí)將高效液相色譜法與現(xiàn)行標(biāo)準(zhǔn)微生物檢測(cè)法進(jìn)行了比較,進(jìn)一步證明高效液相色譜法(HPLC)操作簡(jiǎn)便、快速、精準(zhǔn)的特點(diǎn),為其質(zhì)量控制提供了更為可靠的依據(jù)。
