仿生消化法測定豬飼料原料還原糖釋放量的重復(fù)性和可加性研究

【摘要】:本試驗(yàn)旨在建立單胃動(dòng)物仿生消化系統(tǒng)模擬豬飼料原料消化后還原糖釋放量的測定方法,為飼料養(yǎng)分生物學(xué)效價(jià)的評(píng)估提供參考。還原糖釋放量與上樣量的線性關(guān)系研究以玉米-豆粕混合物(75%玉米+25%豆粕)為研究對(duì)象,上樣量設(shè)0.2、0.4、0.6、0.8和1.0 g 5個(gè)處理,每個(gè)處理4個(gè)重復(fù),每個(gè)重復(fù)1根消化管;方法的重復(fù)性檢驗(yàn)以玉米-豆粕混合物、大麥、花生粕和米糠為研究對(duì)象,每個(gè)樣品設(shè)3個(gè)批次,每個(gè)批次4個(gè)重復(fù);方法的可加性檢驗(yàn)設(shè)19個(gè)處理,其中處理1~7分別為玉米、大麥、高粱、大豆粕、花生粕、棉籽粕和米糠,處理8~19為2種或2種以上飼料原料按不同比例組合制備的12種混合飼料,每個(gè)處理4個(gè)重復(fù),每個(gè)重復(fù)1根消化管,在仿生消化系統(tǒng)中模擬豬消化后測定各處理的還原糖釋放量。結(jié)果表明:上樣量在0.2~0.8 g時(shí),還原糖總釋放量與上樣量呈顯著的線性關(guān)系(R2=0.999 2),還原糖相對(duì)釋放量在559.56~582.70 mg/g DM變化,變異系數(shù)為1.66%,而上樣量為1.0 g時(shí),還原糖相對(duì)釋放量比上樣量為0.2~0.8 g時(shí)的平均值下降5.37%;3個(gè)批次的大麥、花生粕、米糠和玉米-豆粕混合物的還原糖釋放量的批內(nèi)變異系數(shù)、批間變異系數(shù)和總變異系數(shù)均不大于1.68%,批間最大相對(duì)偏差分別為0.68%、1.50%、1.39%和0.29%;12種混合飼料還原糖釋放量的實(shí)測值顯著高于計(jì)算值(P0.05),而還原糖釋放量的計(jì)算值與實(shí)測值的線性回歸模型與y=x相重合(截距P=0.480 5;斜率P=0.514 1)。由此得出,當(dāng)上樣量在0.2~0.8 g時(shí),上樣量與還原糖釋放量呈顯著線性關(guān)系;仿生消化法測定豬飼料原料還原糖釋放量的重復(fù)性和可加性滿足定量分析的基本要求。
[Abstract]:The purpose of this study was to establish a method for the determination of reducing sugar release from pig feed after digestion by bionic digestive system of monogastric animals, and to provide a reference for the evaluation of the biological titer of feed nutrients. The linear relationship between reduced sugar release and sampling volume was studied by using corn-soybean meal mixture (75% corn 25% soybean meal) as the research object. The sample size was 0.2, 0.4, 0.6, 0.8 and 1.0 g, respectively, with 4 repeats in each treatment. One digestive tube per repetition; The reproducibility test of corn-soybean meal mixture, barley, peanut meal and rice husk was carried out. Each sample was divided into 3 batches and 4 repeats per batch. The additivity of the method was determined by 19 treatments, including corn, barley, sorghum, soybean meal, peanut meal, cottonseed meal and rice husk, respectively. Treatment 8 / 19 is 12 kinds of mixed feed prepared by combination of 2 or more feed materials in different proportion, with 4 repeats per treatment and 1 digestive tube per repetition.
