【摘要】：[目的]鹿流行性出血熱是由庫(kù)蠓等媒介昆蟲(chóng)吸血叮咬傳播鹿流行性出血熱病毒引起的一種非接觸性病毒性傳染病,該病毒屬于環(huán)狀病毒屬,呼腸孤病毒科,主要感染野生和家養(yǎng)動(dòng)物。本研究課題主要是為了了解設(shè)立在山西省陽(yáng)城縣藍(lán)舌病監(jiān)控牛場(chǎng)和羊場(chǎng)的監(jiān)控動(dòng)物蟲(chóng)媒病毒感染情況,因此,于2013年5月到2014年6月期間,在山西省陽(yáng)城縣采集監(jiān)控動(dòng)物牛羊的血清、肝素抗凝血對(duì)蟲(chóng)媒病毒進(jìn)行分離和鑒定。[方法]采用競(jìng)爭(zhēng)酶聯(lián)免疫吸附(C-ELISA)試驗(yàn)分別檢測(cè)牛羊血清中的BTV和EHDV抗體,采用微量中和試驗(yàn)檢測(cè)牛羊血清中病毒抗體進(jìn)行病毒型特異性確定,采用細(xì)胞培養(yǎng)、RT-PCR和測(cè)序分析,對(duì)病毒進(jìn)行分離和鑒定。[結(jié)果]在無(wú)臨床癥狀的監(jiān)控牛血液中分離到1株陽(yáng)性分離物,株陽(yáng)性分離物只在BHK-21細(xì)胞上產(chǎn)生規(guī)律性細(xì)胞病變,在C6/36細(xì)胞上不會(huì)出現(xiàn)CPE;對(duì)這株陽(yáng)性分離物細(xì)胞液過(guò)濾后測(cè)其TCID50,滴度分別為104.5TCID50/100μL,采用環(huán)狀病毒屬特異性引物RT-PCR擴(kuò)增檢測(cè),與陽(yáng)性對(duì)照擴(kuò)增片段相似；采用隨機(jī)RT-PCR進(jìn)行擴(kuò)增測(cè)序,將這株病毒的VP2和VP7序列在NBCI上進(jìn)行BLAST比對(duì),同源性達(dá)到99.0%；微量中和試驗(yàn)結(jié)果和分子生物學(xué)試驗(yàn)確定該毒株的血清學(xué)為EHDV-1。[結(jié)論]本次研究首次從山西省的黃牛血液中分離到鹿流行性出血熱病毒,為今后山西省蟲(chóng)媒病毒的分離與鑒定提供一個(gè)重要的參考,為蟲(chóng)媒病毒的防治奠定了理論基礎(chǔ)。
[Abstract]:[objective] Deer epidemic hemorrhagic fever (EHF) is a non-contact viral infectious disease caused by blood sucking and bites by vector insects such as Kucoides. The virus belongs to the genus annular virus and the family Enterovirus. Mainly infected with wild and domestic animals. The main purpose of this study is to understand the monitoring of animal arbovirus infection in cattle and sheep farms with blue tongue disease in Yangcheng County, Shanxi Province. therefore, from May 2013 to June 2014, The serum of monitored animal cattle and sheep was collected in Yangcheng County, Shanxi Province, and heparin anticoagulant was used to isolate and identify the arbovirus. [methods] BTV and EHDV antibodies in bovine sheep serum were detected by competitive enzyme-linked immunosorbent assay (C-ELISA), viral antibodies in bovine sheep serum were determined by microneutralization test, and cell culture was used. RT-PCR and sequencing analysis were used to isolate and identify the virus. [results] A positive isolate was isolated from bovine blood without clinical symptoms. The positive isolate only produced regular cytopathic effect on BHK-21 cells, but no CPE; appeared on C636 cells. The TCID50, titers of the positive isolate were 100 渭 L 104.5TCID50/ after filtration, and the amplified fragments were similar to those of the positive control by RT-PCR amplification with annular virus specific primers. Random RT-PCR was used to amplify and sequence the VP2 and VP7 sequences of the virus on NBCI, and the homology was 99.0%. The serological results of microneutralization test and molecular biology test confirmed that the serology of the strain was EHDV-1.. [conclusion] Deer epidemic hemorrhagic fever virus was isolated from the blood of yellow cattle in Shanxi Province for the first time, which provides an important reference for the isolation and identification of arbovirus in Shanxi Province in the future, and lays a theoretical foundation for the prevention and control of arbovirus.
