一株豬丹毒桿菌分離株的致病性與耐藥性分析

【摘要】:從某豬場病死豬心血、肝、脾等組織分離到1株革蘭陽性桿菌,經(jīng)細菌形態(tài)、16S rRNA基因擴增,確定分離菌為豬丹毒桿菌。對分離菌進一步開展了藥物敏感性試驗、耐藥基因的檢測、細菌致病性試驗及免疫保護性試驗。結(jié)果發(fā)現(xiàn):分離株對β-內(nèi)酰胺類、大環(huán)內(nèi)酯類、氟苯尼考高敏,但對氨基糖苷類、四環(huán)素、林可霉素、諾氟沙星等耐藥;分離株可擴增出氨基糖苷類耐藥基因aac C2和四環(huán)素耐藥基因tetB,但菌株中沒有擴增到四環(huán)素耐藥基因tetA。細菌致病性試驗表明該毒株對小鼠的LD50為1.77×10~5cfu/mL。小鼠免疫保護試驗結(jié)果表明,疫苗G4T10免疫后28 d,以分離株攻毒,對照組全部死亡(5/5),免疫組小鼠攻毒后沒有死亡,僅表現(xiàn)為體重減輕,表明現(xiàn)用豬丹毒疫苗能保護小鼠抵抗豬丹毒的致死性攻擊。
[Abstract]:A strain of Gram-positive bacillus was isolated from the heart blood, liver, spleen and other tissues of dead pigs in a pig farm. After bacterial morphology and 16s rRNA gene amplification, the isolated bacteria were identified as Paeonia viridis. The drug sensitivity test, drug resistance gene detection, bacterial pathogenicity test and immune protection test were further carried out. The results showed that the isolates were resistant to 尾-lactam, macrolides, florfenicol Gao Min, but resistant to aminoglycosides, tetracycline, lincomycin and norfloxacin. The aminoglycoside resistance gene aac C2 and tetracycline resistant gene tetB, could be amplified by the isolate, but the tetracycline resistance gene tetA. was not amplified in the strain. The bacterial pathogenicity test showed that the LD50 of the strain to mice was 1.77 脳 10 ~ 5cfu / mL.
