黔北麻羊繁殖相關基因mRNA表達水平與多態(tài)性分析

【摘要】:本研究以黔北麻羊為主要研究對象,分析FSHβ、FSHR、Gn RH和Gn RHR基因多態(tài)性與繁殖力的關系,Gn RHR和FSHR在發(fā)情間期和發(fā)情期的表達規(guī)律,Gn RHR基因CDS區(qū)功能及結構。本試驗旨在研究黔北麻羊繁殖相關基因多態(tài)與表達在低產(chǎn)和高產(chǎn)山羊的差異,為從分子遺傳方法上提高山羊繁殖力提供科學依據(jù)。研究結果如下:1、FSHβ和Gn RH基因中沒有檢測出多態(tài)位點,均不具有多態(tài)性。2、FSHR基因外顯子1的C126T和第10外顯子的C1246A和C1412A發(fā)現(xiàn)3個SNP位點。遺傳變異與產(chǎn)羔數(shù)關聯(lián)分析,C1246A位點,黔北麻羊個體產(chǎn)羔數(shù)存在差異,C1412A位點黔北麻羊和貴州黑山羊個體產(chǎn)羔數(shù)存在差異;在Gn RHR基因第1外顯子發(fā)現(xiàn)G121A突變。分析發(fā)現(xiàn),在黔北麻羊和貴州黑山羊兩個群體個體的產(chǎn)羔數(shù)存在明顯差異。3、在發(fā)情間期,黔北麻羊BB基因型個體下丘腦和子宮FSHR的表達量高于AA基因型個體(P㩳0.05);黔北麻羊AA基因型垂體、輸卵管、卵巢和子宮Gn RHR的表達量高于GG基因型黔北麻羊(P㩳0.05)。在發(fā)情期,黔北麻羊BB基因型個體卵巢和子宮FSHR的表達量高于AA基因型個體(P㩳0.05);黔北麻羊AA基因型垂體、輸卵管、卵巢和子宮Gn RHR的表達量高于GG基因型黔北麻羊(P㩳0.05)。4、在黔北麻羊垂體中成功克隆Gn RHR基因CDS區(qū)984bp,可編碼328個氨基酸。
[Abstract]:In this study, Guizhou Ma sheep was used as the main research object to analyze the relationship between FSH 尾, FSHR,Gn RH and Gn RHR gene polymorphism and fecundity.
