LiCl對絨山羊毛囊生長和β-catenin表達的影響

【摘要】:為研究不同劑量LiCl對絨山羊毛囊生長和β-連環(huán)蛋白(β-catenin)m RNA表達的影響,將分離的絨山羊初級毛囊隨機分為5組,每組24個重復(fù),對照組為William'E基礎(chǔ)培養(yǎng)液。試驗組在基礎(chǔ)培養(yǎng)液中依次添加2,5,10和20mmol·L~(-1)的LiCl,培養(yǎng)期為7d,每日測量毛囊生長長度。運用熒光定量PCR技術(shù)測定各組β-cateninm RNA表達水平。結(jié)果表明:2~10 mmol·L~(-1)LiCl組的β-catenin m RNA表達水平和生長長度極顯著高于對照組(p0.01),毛囊形態(tài)維持良好;LiCl濃度為20mmol·L~(-1)組的β-catenin m RNA表達量及生長長度極顯著低于對照組(p0.01),體外培養(yǎng)毛囊生長受到抑制。添加2~10mmol·L~(-1)的LiCl可促進絨山羊毛囊生長,促使β-catenin m RNA表達水平提高,其中5mmol·L~(-1)為較優(yōu)濃度。通過本試驗研究發(fā)現(xiàn),適量的LiCl在一定程度上有利于初級毛囊的生長及形態(tài)的維持,促進了β-catenin m RNA表達。
