FSH及Wnt信號通路對綿羊卵泡顆粒細胞的增殖及雌激素分泌的影響

【摘要】:旨在研究Wnt信號通路對綿羊卵泡顆粒細胞增殖及雌激素分泌的影響,探究其是否與促卵泡素(Follicular stimulating hormone,FSH)有關(guān)。在添加有FSH以及不同濃度Wnt信號通路抑制劑IWR-1的培養(yǎng)基中培養(yǎng)綿羊卵巢卵泡顆粒細胞168h,利用Guava ViaCount?Reagent測定細胞數(shù)目的變化,利用綿羊ELISA試劑盒檢測雌激素濃度的變化,利用qRT-PCR檢測FSH靶基因CYP19和CCND2的相對表達量差異。結(jié)果表明,無論有無FSH,抑制劑IWR-1使顆粒細胞數(shù)目顯著減少(P0.05);當有FSH時,抑制劑IWR-1的添加導致雌激素濃度顯著降低(P0.05)。IWR-1的濃度為1.0μmol·L-1時,對顆粒細胞的抑制效果最明顯。IWR-1還導致CYP19和CCND2mRNA的表達量降低。綜上表明,Wnt信號通路對綿羊卵泡顆粒細胞的生物學功能具有促進作用,并且這種促進作用可能與FSH有關(guān)。
[Abstract]:The aim of this study was to investigate the effect of Wnt signaling pathway on the proliferation and estrogen secretion of ovine follicular granulosa cells, and to explore whether it is related to follicle stimulating hormone (Follicular stimulating hormone,FSH). Ovine follicular granulosa cells were cultured in the medium supplemented with FSH and different concentration of Wnt signaling pathway inhibitor IWR-1 for 168 hours. The changes of cell number and estrogen concentration were measured by Guava ViaCount?Reagent and ELISA kit, respectively. The relative expression of FSH target gene CYP19 and CCND2 was detected by qRT-PCR. The results showed that the number of granulosa cells was significantly decreased with or without FSH, inhibitor IWR-1 (P0.05). At the concentration of 1.0 渭 mol 路L-1 of IWR-1, the inhibitory effect on granulosa cells was the most obvious. The expression of CYP19 and CCND2mRNA was also decreased by IWR-1 when the concentration of FSH was 1.0 渭 mol 路L-1. The concentration of estrogens decreased significantly with the addition of IWR-1 (P0.05). The results indicated that Wnt signaling pathway could promote the biological function of ovine follicular granulosa cells, and this effect might be related to FSH.
