腦紅蛋白在西門塔爾牛主要器官組織中分布的研究

【摘要】:為了探尋腦紅蛋白(Neuroglobin,NGB)在成年西門塔爾牛不同組織細(xì)胞的分布情況,選擇健康成年西門塔爾牛5只,利用免疫組織化學(xué)染色SP法觀察NGB在成年西門塔爾牛不同組織細(xì)胞的分布特征,結(jié)果顯示:NGB在已檢測(cè)的多數(shù)組織細(xì)胞中均有陽(yáng)性表達(dá),不同個(gè)體之間的NGB表達(dá)無(wú)明顯差異。中樞神經(jīng)系統(tǒng)中,NGB免疫陽(yáng)性反應(yīng)主要定位于神經(jīng)細(xì)胞。大腦灰質(zhì)中NGB陽(yáng)性細(xì)胞廣泛分布于大腦灰質(zhì)第I至第VI層,免疫陽(yáng)性反應(yīng)強(qiáng)度在大腦灰質(zhì)第I~VI層之間沒有明顯的差異,大腦白質(zhì)區(qū)域也可見散在的NGB陽(yáng)性細(xì)胞。嗅球的僧帽細(xì)胞層NGB免疫陽(yáng)性反應(yīng)相對(duì)強(qiáng),胞體大。海馬不同區(qū)域的錐體細(xì)胞中均可見NGB陽(yáng)性表達(dá)。小腦灰質(zhì)各層細(xì)胞中廣泛分布有NGB陽(yáng)性細(xì)胞,其中浦肯野細(xì)胞層免疫陽(yáng)性反應(yīng)強(qiáng)于顆粒層和分子層細(xì)胞,小腦白質(zhì)中未見明顯的陽(yáng)性反應(yīng)。在脊髓各區(qū)段,NGB陽(yáng)性細(xì)胞主要分布于灰質(zhì),白質(zhì)中陽(yáng)性反應(yīng)相對(duì)較弱。內(nèi)分泌系統(tǒng)中,腎上腺皮質(zhì)和髓質(zhì)細(xì)胞均呈陽(yáng)性反應(yīng),皮質(zhì)中球狀帶細(xì)胞陽(yáng)性反應(yīng)最強(qiáng),網(wǎng)狀帶次之,束狀帶最弱,髓質(zhì)中反應(yīng)普遍較皮質(zhì)弱。腺垂體內(nèi)腺細(xì)胞可見NGB陽(yáng)性表達(dá),表達(dá)部位主要在細(xì)胞質(zhì),神經(jīng)垂體內(nèi)神經(jīng)纖維亦呈陽(yáng)性反應(yīng)。松果體中可見有廣泛分布的NGB陽(yáng)性細(xì)胞。在雄性生殖系統(tǒng)中,睪丸各級(jí)精母細(xì)胞和部分間質(zhì)細(xì)胞均顯示有NGB陽(yáng)性表達(dá),附睪輸出小管和附睪管上皮細(xì)胞內(nèi)亦可見NGB陽(yáng)性表達(dá),輸精管未見表達(dá)。雌性生殖系統(tǒng)中,卵巢原始卵泡細(xì)胞及各級(jí)生長(zhǎng)卵泡細(xì)胞的顆粒層中均有NGB陽(yáng)性反應(yīng),子宮內(nèi)膜上皮細(xì)胞和子宮腺導(dǎo)管上皮細(xì)胞呈NGB強(qiáng)陽(yáng)性表達(dá)。成年西門塔爾牛消化系統(tǒng)中,多室胃前胃黏膜上皮細(xì)胞中均可見較強(qiáng)的NGB陽(yáng)性反應(yīng),各腸段腸腺細(xì)胞中均有較弱的NGB陽(yáng)性反應(yīng),但在十二指腸段的小腸腺細(xì)胞較十二指腸腺細(xì)胞的陽(yáng)性表達(dá)強(qiáng)。肝臟膽小管可見有NGB陽(yáng)性反應(yīng)。胰腺中胰島細(xì)胞及腺泡細(xì)胞均可見NGB陽(yáng)性表達(dá),表達(dá)部位主要在細(xì)胞質(zhì),其中胰島細(xì)胞陽(yáng)性反應(yīng)較強(qiáng)。頜下腺的紋狀管上皮細(xì)胞有NGB陽(yáng)性表達(dá)。腮腺腺泡細(xì)胞和導(dǎo)管也均有較弱的NGB陽(yáng)性反應(yīng)。西門塔爾牛其他組織細(xì)胞,心肌纖維中可見較弱的NGB陽(yáng)性表達(dá)。脾臟的紅髓中可見NGB陽(yáng)性細(xì)胞,白髓中未見表達(dá)。肺臟細(xì)支氣管上皮細(xì)胞及肺泡上皮細(xì)胞中均可見較弱的NGB陽(yáng)性表達(dá),表達(dá)部位主要在胞質(zhì)。腎臟的遠(yuǎn)曲小管、近曲小管和集合小管的上皮細(xì)胞均有較弱的NGB陽(yáng)性表達(dá),腎小球內(nèi)未見有明顯的NGB陽(yáng)性反應(yīng)。膀胱中未見NGB陽(yáng)性反應(yīng)。NGB在成年西門塔爾牛多種器官組織細(xì)胞中的廣泛分布,提示NGB不僅在氧的利用及功能活動(dòng)中發(fā)揮作用,在其它生理活動(dòng)中可能也擔(dān)負(fù)著重要的功能。
[Abstract]:In order to investigate the distribution of brain erythrin (Neuroglobin,NGB) in different tissues and cells of adult Simmental cattle, five healthy adult Simmental cattle were selected. The distribution of NGB in different tissues and cells of adult Simmental cattle was observed by immunohistochemical staining SP method. The results showed that NGB was positive in most of the detected tissues and cells, and there was no significant difference in the expression of NGB among different individuals. In the central nervous system, NGB immunoreactivity was mainly localized in nerve cells. The NGB positive cells in the gray matter were widely distributed in the layers I to VI of the gray matter. There was no significant difference in the intensity of the immunoreactive reaction between the layers I ~ VI in the gray matter, and the scattered NGB positive cells could also be seen in the white matter area of the brain. The NGB immunoreactivity in the cell layer of olfactory bulb was relatively strong and the cell body was large. The positive expression of NGB was found in pyramidal cells in different regions of hippocampus. NGB positive cells were widely distributed in all layers of cerebellar gray matter. The immunopositive reaction in Purkinje cell layer was stronger than that in granular layer and molecular layer, and there was no obvious positive reaction in cerebellar white matter. In all segments of spinal cord, NGB positive cells were mainly distributed in gray matter, while the positive reaction in white matter was relatively weak. In the endocrine system, the adrenal cortex and medulla cells were positive, the globular zone cells in the cortex were the strongest, the reticular zone was the second, the fascicular zone was the weakest, and the reaction in the medulla was generally weaker than that in the cortex. The positive expression of NGB was observed in adenohypophysis cells, mainly located in cytoplasm and nerve fibers in neurohypophysis. A wide range of NGB positive cells were found in pineal gland. In male reproductive system, the positive expression of NGB was observed in spermatocytes and some Leydig cells of testis. The positive expression of NGB was also observed in epididymal output tubule and epididymal duct epithelial cells, and there was no expression in vas deferens. In female reproductive system, NGB positive reaction was found in ovarian primordial follicular cells and granular layers of all grade growing follicular cells, and strong positive expression of NGB was observed in endometrial epithelial cells and glandular ductal epithelial cells of uterus. In the digestive system of adult Simmental cattle, strong positive reaction of NGB was observed in the epithelial cells of multiloculed forestomach mucosa, and weak positive reaction of NGB was found in the intestinal gland cells of each intestinal segment. However, the positive expression of small intestinal gland cells in duodenum segment was stronger than that in duodenum gland cells. The positive reaction of NGB was found in the bile duct of the liver. The positive expression of NGB was found in pancreatic islet cells and acinar cells, mainly in cytoplasm, and the positive reaction of pancreatic islet cells was stronger than that of pancreatic acinar cells. There was positive expression of NGB in the striate duct epithelial cells of submandibular gland. Parotid acinar cells and ductus were also weakly positive for NGB. In other tissues and cells of Simmental cattle, weak positive expression of NGB was found in myocardial fibers. NGB positive cells were found in the red pulp of spleen, but no expression was found in the white pulp. Weak positive expression of NGB was found in bronchiolar epithelial cells and alveolar epithelial cells, mainly in cytoplasm. The epithelial cells of distal convoluted tubule, proximal convoluted tubule and collecting tubule all showed weak positive expression of NGB, but no obvious positive reaction of NGB was found in glomeruli. There was no NGB positive reaction in bladder. The extensive distribution of NGB in many organs, tissues and cells of adult Simmental cattle suggested that NGB not only played an important role in oxygen utilization and functional activities, but also played an important role in other physiological activities.
