性別相關(guān)基因在牛早期性別發(fā)育中的表達(dá)規(guī)律研究

【摘要】:旨在研究SRY、SOX9、DAX1和FOXL2 4個(gè)性別相關(guān)基因在早期牛胎兒中的表達(dá)情況,闡明其表達(dá)規(guī)律。收集34~80胎齡(dpc)牛早期胎兒的生殖嵴和中腎,先確定其性別,然后采用熒光定量PCR和免疫組化方法分別對SRY、SOX9、DAX1、FOXL2基因在RNA和蛋白水平進(jìn)行定量和定位檢測。經(jīng)4對引物的性別鑒定,19個(gè)胎兒中10個(gè)為雄性,9個(gè)為雌性。SRY基因只在雄性中表達(dá),而FOXL2基因在雌性中有顯著表達(dá),在雄性中則有痕量的表達(dá)。在雄性生殖嵴中,SOX9和DAX1的表達(dá)模式相似,但在44~56dpc DAX1表達(dá)量高于SOX9;在38~80dpc的雌性生殖嵴中,DAX1的表達(dá)量持續(xù)高于SOX9,且53dpc后,SOX9表達(dá)量持續(xù)維持低水平,而DAX1表達(dá)量卻急劇升高到80dpc?梢,牛早期胎兒雄性的性別決定時(shí)間為35~39dpc,雌性則為39~41dpc;且SRY和FOXL2分別為牛早期胎兒雄性和雌性的性別決定基因,本研究初步揭示了牛早期胎兒性別相關(guān)基因的表達(dá)規(guī)律,為家畜性別控制提供理論基礎(chǔ)。
[Abstract]:The purpose of this study was to investigate the expression of four sex-related genes SRY,SOX9,DAX1 and FOXL2 in early bovine fetuses. The reproductive crest and mesonephros of early fetuses of 34-80 gestational age (dpc) cattle were collected and their sex was determined. Then quantitative and localization of SRY,SOX9,DAX1,FOXL2 gene in RNA and protein levels were detected by fluorescence quantitative PCR and immunohistochemical method respectively. According to sex identification of 4 pairs of primers, 10 of 19 fetuses were male and 9 were female. SRY gene was expressed only in male, while FOXL2 gene was significantly expressed in female and trace expression in male. In male genital crest, the expression patterns of SOX9 and DAX1 were similar, but the expression of 44~56dpc DAX1 was higher than that of SOX9;. In the female genital crest of 38~80dpc, the expression level of DAX1 was higher than that of SOX9, and after 53dpc, the expression of SOX9 remained low, while the expression of DAX1 increased sharply to 80 dpc. It can be seen that the sex-determining time of male and female fetuses is 35-39 dpcs and 39-41-dpcs respectively. SRY and FOXL2 are male and female sex-determining genes of early bovine fetus, respectively. This study revealed the expression rule of bovine early fetal sex-related genes, and provided a theoretical basis for sex control of domestic animals.
