低劑量多巴胺對肉雞腎臟鈉鉀氯共轉運體表達的影響

【摘要】:為研究低劑量多巴胺對肉雞腎臟鈉鉀氯共轉運體(NKCC2)表達的影響,按常規(guī)條件飼養(yǎng)AA肉雞40羽,25日齡時隨機分為對照組和試驗組,對照組肉雞經(jīng)靜脈注射5μg/kg/min 0.9%生理鹽水,試驗組肉雞按相同方法注射等體積低劑量左旋多巴胺,并采用組織形態(tài)學觀察、血尿生化檢測、蛋白質免疫印跡試驗和RT-q PCR等方法檢測腎組織形態(tài)、各生化指標的變化、NKCC2蛋白水平和mRNA水平的變化。結果顯示:與對照組相比,試驗組在給予低劑量左旋多巴胺后,尿素氮(BUN)、肌酐(Cr)及平均動脈壓(m PAP)水平均未發(fā)生明顯改變(P0.05);低劑量左旋多巴胺持續(xù)給藥后,肉雞腎小管上皮細胞胞膜NKCC2蛋白水平顯著降低(P0.05),細胞質NKCC2蛋白顯著升高(P0.05);然而NKCC2 mRNA水平未發(fā)生明顯變化(P0.05)。結論:在不影響腎小球濾過率及血壓前提下,低劑量多巴胺能夠引起肉雞機體持續(xù)的利鈉利尿作用,其機制與NKCC2蛋白的穿梭作用有關。
[Abstract]:In order to study the effect of low dose dopamine on the expression of sodium, potassium and chloride cotransporter (NKCC2) in kidney of broilers, 40 AA broilers were fed with conventional conditions and randomly divided into control group and experimental group at the age of 25 days. In the control group, 5 渭 g / kg/min 0.9% saline was injected intravenously, and the same volume low dose of L-dopamine was injected into the broilers of the experimental group by the same method. The histological observation and biochemical examination of hematuria were performed in the experimental group. Western blot and RT-q PCR were used to detect the changes of renal tissue morphology, biochemical indexes, NKCC2 protein level and mRNA level. The results showed that compared with the control group, the levels of urea nitrogen (BUN), creatinine (Cr) and mean arterial pressure (m PAP) did not change significantly in the experimental group after low dose of L-dopamine (P0.05). After continuous administration of low dose of L-dopamine, the level of membrane NKCC2 protein in renal tubular epithelial cells of broilers was significantly decreased (P0.05), and the level of cytoplasmic NKCC2 protein was significantly increased (P0.05), but the level of NKCC2 mRNA did not change significantly (P0.05). Conclusion: low dose dopamine can induce continuous natriuretic diuresis in broilers without affecting glomerular filtration rate and blood pressure, and the mechanism is related to the shuttle effect of NKCC2 protein.
