【摘要】：近年來(lái)clade2.3.2.1c中的H5N1亞型禽流感病毒的抗原性發(fā)生了很大變化,Re-6疫苗并不能對(duì)其提供有效保護(hù)。為對(duì)該分支毒株進(jìn)行有效防控,研究以A/Puerto Rico/8/34(H1N1,PR8)毒株為內(nèi)部基因供體,以clade2.3.2.1c的H5N1亞型毒株A/chicken/Jiangsu/YB7/2015(YB7)的HA和NA基因作外部供體,并刪除YB7株HA中裂解位點(diǎn)處的多個(gè)堿性氨基酸,通過反向遺傳方法成功拯救出1株重組病毒r YB7。r YB7株病毒在雞胚和MDCK細(xì)胞上均有較好的繁殖性能,對(duì)SPF雞胚和雞均無(wú)致病性。本實(shí)驗(yàn)室在對(duì)毒株抗原性研究的基礎(chǔ)上研發(fā)的疫苗候選株為防控變異的clade2.3.2.1c的禽流感病毒提供了良好的疫苗備選。
[Abstract]:In recent years, the antigenicity of H5N1 subtype avian influenza virus (AIV) in clade2.3.2.1c has changed greatly, and Re-6 vaccine can not provide effective protection. In order to prevent and control the strain effectively, A/Puerto Rico/8/34 (H1 N 1, PR8) strain was used as the internal gene donor. The HA and NA genes of A/chicken/Jiangsu/YB7/2015 (YB7) of clade2.3.2.1c subtype strain A/chicken/Jiangsu/YB7/2015 (YB7) were used as external donors, and several basic amino acids at the cleavage site of YB7 strain HA were deleted. A recombinant virus r YB7.r YB7 strain was successfully rescued by reverse genetic method. It had good reproductive performance on both chicken embryos and MDCK cells, and had no pathogenicity to both SPF chicken embryos and chickens. The vaccine candidate strain developed by our laboratory based on the study of the antigenicity of the strain provides a good vaccine option for the prevention and control of the avian influenza virus (AIV) of the mutated clade2.3.2.1c.
