一株產(chǎn)纖維素酶的解淀粉芽孢桿菌的分離鑒定及其酶促反應(yīng)適宜條件初步研究

【摘要】:為了分離、篩選產(chǎn)纖維素酶益生菌,確定其酶促反應(yīng)適宜條件,利用羧甲基纖維素鈉等作為篩選培養(yǎng)基,結(jié)合剛果紅染色法,從玉米青貯飼料樣品中篩選產(chǎn)纖維素酶益生菌,并通過形態(tài)學(xué)和系統(tǒng)進化樹方法鑒定分離菌。同時對培養(yǎng)時間、pH和溫度等酶促反應(yīng)條件進行了研究。結(jié)果表明,篩選出1株可降解羧甲基纖維素,并產(chǎn)生清晰透明圈的菌株。經(jīng)形態(tài)學(xué)觀察和16SrRNA基因序列分析,鑒定該菌為解淀粉芽孢桿菌(Bacillus amyloliquefaciens)。粗酶液中濾紙酶活為1.5U/mL,外切葡聚糖酶活為1.3U/mL,內(nèi)切葡聚糖酶活為6.4U/mL,β-葡萄糖苷酶活為2.3U/mL。酶促反應(yīng)的適宜條件為pH 5.5,溫度50℃,粗酶液為培養(yǎng)3d后發(fā)酵液上清。解淀粉芽孢桿菌分離株具有一定的產(chǎn)纖維素酶能力,在玉米秸稈生物降解與利用方面具有潛在的開發(fā)價值。
[Abstract]:In order to isolate and screen cellulase-producing probiotics and determine the suitable conditions for enzymatic reaction, cellulase-producing probiotics were screened from corn silage samples by using carboxymethyl cellulose sodium as screening medium and Congo red staining method. The isolated bacteria were identified by morphological and phylogenetic tree methods. At the same time, the conditions of enzymatic reaction, such as culture time, pH and temperature, were studied. The results showed that a strain which could degrade carboxymethyl cellulose and produce clear transparent circle was screened out.
