【摘要】：為探究禽流感病毒(AIV)M2蛋白對(duì)細(xì)胞自噬的影響,本研究構(gòu)建了pEGFP-LC3和pCAF-M2重組質(zhì)粒,并采用脂質(zhì)體將這兩種重組表達(dá)質(zhì)粒共轉(zhuǎn)染293T細(xì)胞或Hela細(xì)胞中,同時(shí)以單獨(dú)轉(zhuǎn)染這兩種重組質(zhì)粒的細(xì)胞作為對(duì)照組,利用間接免疫熒光和westernblot的方法檢測(cè)細(xì)胞自噬體及自噬流情況。自噬體檢測(cè)結(jié)果表明,AIVM2蛋白能夠誘導(dǎo)轉(zhuǎn)染細(xì)胞內(nèi)自噬體積累。通過LC3-IIturnover試驗(yàn)、mRFP-GFP-LC3雙熒光指示系統(tǒng)以及pEGFP-LC3與LAMP1的共定位檢測(cè)進(jìn)一步證明,M2蛋白通過阻斷自噬體與溶酶體融合,抑制溶酶體對(duì)自噬體的降解從而誘導(dǎo)自噬體積累。本研究揭示AIVM2蛋白對(duì)細(xì)胞自噬的發(fā)生具有重要作用,為深入研究自噬在A型流感病毒復(fù)制中的調(diào)節(jié)作用奠定基礎(chǔ)。
[Abstract]:In order to investigate the effect of avian influenza virus (AIV) M2 protein on autophagy, recombinant plasmids of pEGFP-LC3 and pCAF-M2 were constructed and co-transfected into 293T cells or Hela cells by lipofectamine. At the same time, the autophagy and autophagy were detected by indirect immunofluorescence and westernblot in cells transfected with these two recombinant plasmids as control group. Autophagy assay showed that AIVM2 protein could induce the accumulation of autophagy in transfected cells. LC3-IIturnover test, mRFP-GFP-LC3 double fluorescence indicator system and co-localization of pEGFP-LC3 and LAMP1 further demonstrated that M2 protein could block the fusion of autophagy and lysosome. Inhibition of lysosome degradation of autophagy and induction of autophagy accumulation. This study reveals that AIVM2 protein plays an important role in the occurrence of autophagy and lays a foundation for further study on the regulation of autophagy in the replication of influenza A virus.
