豬繁殖與呼吸綜合征病毒感染豬肺泡巨噬細胞天然免疫信號通路相關分子變化分析

【摘要】:利用蛋白質(zhì)組學的方法對豬繁殖與呼吸綜合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)感染豬肺泡巨噬細胞(pulmonary macrophage,PAM)后的蛋白質(zhì)動態(tài)變化進行探討,為闡明病毒感染后引起的復雜免疫學反應及為該病的防控提供理論依據(jù)。通過Label-free LC-MS/MS方法定量鑒定PRRSV感染PAMs后差異表達細胞蛋白質(zhì)。結(jié)果顯示,共鑒定出430個顯著差異表達蛋白質(zhì),其中包括171個上調(diào)表達蛋白質(zhì)和259個下調(diào)表達蛋白質(zhì),這些蛋白質(zhì)與細胞抗病毒天然免疫及抗原遞呈等相關。值得關注的是,許多上調(diào)表達蛋白質(zhì)都富集在與天然免疫相關的NF-κB、MAPK及RIG-Ⅰ等信號通路上,包括STAT1/2、OAS1/2、Mx1/2、ISG15/20、IFIT1/2/3/5等;下調(diào)蛋白質(zhì)主要富集在MHCⅠ和MHCⅡ分子相關信號通路上。熒光定量PCR和Western blot結(jié)果證實,PRRSV感染導致了STAT1/2、OAS1/2、Mx1、CD14、ISG和IFIT等的轉(zhuǎn)錄或表達,這為進一步闡述PRRSV的感染機制奠定基礎。
[Abstract]:The dynamic changes of protein in porcine alveolar macrophages (pulmonary macrophage,PAM) infected with porcine reproductive and respiratory syndrome virus (porcine reproductive and respiratory syndrome virus,PRRSV) were studied by proteomics. In order to clarify the complex immune response caused by virus infection and to provide a theoretical basis for the prevention and control of the disease. Differential expression of cellular proteins after PRRSV infection with PAMs was quantitatively identified by Label-free LC-MS/MS. The results showed that 430 differentially expressed proteins were identified, including 171 up-regulated proteins and 259 down-regulated proteins, which were related to cellular antiviral innate immunity and antigen presentation. It is worth noting that many up-regulated proteins are concentrated in NF- 魏 B map-MAPK and RIG- I signaling pathways related to innate immunity, including STAT1/2,OAS1/2,Mx1/2,ISG15/20,IFIT1/2/3/5.
