【摘要】：由于常規(guī)PCR所使用的加熱機(jī)制為熱傳導(dǎo)的熱循環(huán)儀,需要一個(gè)精密的數(shù)字控制器來快速加熱和冷卻樣品。近年來,越來越多的研究致力于開發(fā)一種快速、便攜式的核酸檢測PCR平臺。而自然對流作為另一種熱轉(zhuǎn)換方式,能促進(jìn)液體自發(fā)反復(fù)循環(huán)以提供工作所需的變性、退火和延伸溫度,在核酸檢測領(lǐng)域有很大的應(yīng)用前景。恒溫?zé)岣艚^式PCR(insulated isothermal PCR,iiPCR)就是采用Rayleigh-Be′nard自然對流原理,利用儀器底部單一熱源對特殊聚碳酸酯毛細(xì)反應(yīng)管底部的紫銅圈接觸加熱,毛細(xì)管上部置入隔熱擋板,上下的溫度差從而使毛細(xì)管內(nèi)部流體對流自發(fā)形成可供PCR擴(kuò)增反應(yīng)的連續(xù)的溫度梯度,LED屏上直接顯示檢測結(jié)果。與傳統(tǒng)的PCR檢測方法相比,具有操作簡便、快速高效、特異性強(qiáng)、儀器成本低等優(yōu)點(diǎn),已開始運(yùn)用于臨床各種病原體的檢測。
[Abstract]:Because the heating mechanism used in conventional PCR is a heat conduction thermometer, a precise digital controller is needed to quickly heat and cool samples. In recent years, more and more research has been devoted to the development of a fast, portable PCR platform for nucleic acid detection. As another way of heat transfer, natural convection can promote the spontaneous repeated circulation of liquid to provide the necessary denaturation, annealing and extension temperature for work. It has great application prospect in the field of nucleic acid detection. Constant temperature and heat isolation type PCR (insulated isothermal PCR,iiPCR) is based on the principle of Rayleigh-Be'nard natural convection, using a single heat source at the bottom of the instrument to heat the copper ring at the bottom of the special polycarbonate capillary tube, and placing the upper part of the capillary tube into an insulating baffle. The temperature difference between the upper and lower levels makes the convection in the capillary tube spontaneously form a continuous temperature gradient for PCR amplification reaction. The detection results are directly displayed on the LED screen. Compared with traditional PCR detection method, it has many advantages, such as simple operation, high efficiency, specificity and low cost of instrument, so it has been applied to the detection of clinical pathogens.
