產(chǎn)腸毒素性大腸桿菌侵染豬腸上皮細(xì)胞后DLG5基因的表達(dá)變化分析

【摘要】:為了揭示DLG5基因在機(jī)體抵抗大腸桿菌刺激過(guò)程中發(fā)揮的功能,本試驗(yàn)利用產(chǎn)腸毒素性大腸桿菌(F18ab、F18ac和K88ac)侵染豬腸上皮細(xì)胞系(IPEC-J2)模擬個(gè)體的致病過(guò)程,用實(shí)時(shí)熒光定量檢測(cè)DLG5基因在感染前后的表達(dá)變化情況,在細(xì)胞水平上探討DLG5基因表達(dá)水平與產(chǎn)腸毒素性大腸桿菌抗性的關(guān)系。結(jié)果表明,3種大腸桿菌的菌清和菌體刺激IPEC-J2后,DLG5基因表達(dá)水平均發(fā)生顯著或極顯著上調(diào);并且由F18ab和K88ac這2種菌體刺激后DLG5基因表達(dá)水平上調(diào)的程度要極顯著高于F18ac菌體刺激后的表達(dá)水平。本研究結(jié)果提示,豬DLG5基因表達(dá)和大腸桿菌的抗性具有密切關(guān)系,可能與其在一定程度上能夠促進(jìn)腸道屏障的結(jié)構(gòu)穩(wěn)定和功能發(fā)揮有關(guān)。
[Abstract]:In order to reveal the function of DLG5 gene in resisting the stimulation of Escherichia coli, we used enterotoxigenic Escherichia coli (F18abanF18ac and K88ac) to infect porcine intestinal epithelial cell line (IPEC-J2) to simulate the pathogenicity of the individual. The expression of DLG5 gene before and after infection was detected by real-time fluorescence quantitative analysis. The relationship between the expression level of DLG5 gene and the resistance of enterotoxigenic Escherichia coli was investigated at the cellular level. The results showed that the level of DLG5 gene expression was significantly up-regulated or extremely significantly up-regulated by the three Escherichia coli strains and the IPEC-J2 stimulated by the bacteria. The expression level of DLG5 gene stimulated by F18ab and K88ac was significantly higher than that stimulated by F18ac. The results suggest that the expression of porcine DLG5 gene is closely related to the resistance of Escherichia coli, and it may be related to the structural stability and function of intestinal barrier.
