【摘要】：采集疑似心包積水-包涵體肝炎綜合征(HPS-IBH)病雞肝臟、肺臟、脾臟、法氏囊等病料,采用針對(duì)禽腺病毒(FAdV)的Fiber基因及傳染性法氏囊病病毒(IBDV)的vVP2、VP1-b基因設(shè)計(jì)特異引物進(jìn)行病毒的PCR檢測(cè)以及基因序列測(cè)定與分析,并對(duì)病變肝臟組織進(jìn)行組織病理學(xué)觀察。結(jié)果:分別在肝臟、脾臟、肺臟組織中擴(kuò)增出FAdV-Fiber基因的目的條帶,在法氏囊和脾臟組織中擴(kuò)增出IBDV-vVP2、-VP1-b基因的目的條帶;經(jīng)測(cè)序分析,該病例中FAdV屬于Ⅰ群禽腺病毒血清4型(FAdV-4),IBDV屬于基因重排病毒;病變肝臟組織H-E染色觀察顯示,肝細(xì)胞核內(nèi)有典型的嗜堿性包涵體。綜合分析,確診該HPS-IBH病例是由于雞群感染IBDV后繼發(fā)感染FAdV-4所致。
[Abstract]:Liver, lung, spleen and bursa were collected from chickens with suspected pericardial hydronephrosis and inclusion body hepatitis syndrome (HPS-IBH). Fiber gene of (FAdV) and vVP2, of (IBDV) of infectious bursal disease virus were used. The specific primers designed for VP1-b gene were used to detect the virus by PCR, to detect the gene sequence, and to observe the histopathology of the diseased liver tissue. Results: the target bands of FAdV-Fiber gene were amplified in liver, spleen and lung, and IBDV-vVP2,-VP1-b gene was amplified in bursa of Fabricius and spleen.
