一周歲內(nèi)灘羊皮膚差異表達(dá)蛋白研究

【摘要】:為研究一周歲內(nèi)灘羊皮膚中的差異表達(dá)蛋白,本試驗(yàn)利用雙向凝膠電泳(2-DE)技術(shù)分離了灘羊不同生長階段皮膚蛋白,經(jīng)考馬斯亮藍(lán)染色,用PDQuest 8.0軟件檢測并分析差異蛋白點(diǎn),并經(jīng)MALDITOF/TOF-MS/MS鑒定。對4個(gè)生長時(shí)期的蛋白圖譜進(jìn)行兩兩比對,共發(fā)現(xiàn)了19個(gè)差異蛋白點(diǎn),質(zhì)譜成功鑒定出15個(gè)蛋白點(diǎn)共13種蛋白,可能與灘羊皮膚生長調(diào)控有關(guān)的蛋白有14-3-3蛋白σ亞型(14-3-3protein sigma)、膜聯(lián)蛋白A2(annexin A2)、角蛋白25(keratin 25)、微管蛋白α鏈(tubulin alpha chain)。對差異表達(dá)蛋白進(jìn)行分析,發(fā)現(xiàn)這些差異表達(dá)蛋白可能與灘羊皮膚不同生長階段的毛囊周期性變化有關(guān)。一周歲內(nèi)灘羊皮膚差異蛋白的發(fā)現(xiàn)為灘羊皮膚的年變化規(guī)律研究奠定理論基礎(chǔ)。
[Abstract]:In order to study the differentially expressed proteins in the skin of Tan sheep within one year old, the skin proteins of Tan sheep at different growth stages were isolated by two-dimensional gel electrophoresis (2-DE) and stained with Coomassie brilliant blue. Differential protein spots were detected and analyzed by PDQuest 8.0 software, and identified by MALDITOF/TOF-MS/MS. A total of 19 differential protein spots were found by pairwise comparison of protein maps of 4 growth stages, and 13 proteins were successfully identified by mass spectrometry. There are 14-3-3 protein 蟽 subtypes (14-3-3protein sigma), binding protein A2 (annexin A2), keratin 25 (keratin 25), tubulin 偽 chain (tubulin alpha chain).) that may be related to the regulation of skin growth in Tan sheep. It was found that these differentially expressed proteins might be related to the periodic changes of hair follicles in different growth stages of Tan sheep skin. The discovery of skin differential protein in Tan sheep within one year of age lays a theoretical foundation for the study of annual variation of Tan sheep skin.
