貓杯狀病毒2280株反向遺傳系統(tǒng)的構(gòu)建

【摘要】:為構(gòu)建貓杯狀病毒(FCV)2280株感染性克隆,本研究在FCV 2280株基因組5'末端和3'末端分別引入T7 RNA聚合酶啟動(dòng)子和丁型肝炎核酶(Hdv Rz)序列;并將FCV基因組中唯一的XbaⅠ限制性酶切位點(diǎn)序列進(jìn)行同義突變作為遺傳標(biāo)記;將基因組分為3段分別進(jìn)行RT-PCR擴(kuò)增,通過酶切依次克隆于pOK-12載體中,構(gòu)建FCV基因組全長cDNA重組質(zhì)粒。以其為模板經(jīng)體外轉(zhuǎn)錄制備其全長基因組RNA,并在5'末端加帽,轉(zhuǎn)染CRFK細(xì)胞,24 h后出現(xiàn)典型的細(xì)胞病變。提取出現(xiàn)細(xì)胞病變的病毒液的總RNA制備cDNA模板,PCR擴(kuò)增FCV遺傳標(biāo)記片段并進(jìn)行XbaⅠ酶切鑒定分析,表明拯救的病毒為FCV重組病毒。而且拯救的病毒生長動(dòng)力學(xué)與親本病毒無明顯差異。本實(shí)驗(yàn)為進(jìn)一步研究FCV的生物學(xué)特性和致病機(jī)制奠定了基礎(chǔ)。
