【摘要】：以表達(dá)EGFP報告基因的重組I型馬立克病毒疫苗株(CVI988/Rispens)rMDV-EGFP為母本病毒,通過同源重組的方法用表達(dá)傳染性喉氣管炎病毒(ILTV)gB和gD基因的表達(dá)盒替換病毒rMDV-EGFP中的EGFP報告基因表達(dá)盒,分別獲得了表達(dá)gB和gD基因的重組馬立克病毒rMDV-ILTgB和rMDV-ILTgD。經(jīng)PCR和間接免疫熒光試驗鑒定,證明重組病毒中插入的目的基因大小正確,且能夠表達(dá)gB和gD目的蛋白。免疫效力試驗結(jié)果表明:與弱毒疫苗相比,重組病毒rMDV-ILTgB和rMDV-ILTgD均可對ILTV強毒攻擊提供更好的免疫保護。重組病毒免疫組試驗雞不僅在發(fā)病率、致死率和攻毒后體重增長等指標(biāo)方面與攻毒對照組均存在顯著差異,而且與常規(guī)弱毒疫苗免疫組在致死率和攻毒后體重增長指標(biāo)方面也存在顯著差異。
[Abstract]:The recombinant Marek virus type I vaccine strain (CVI988/Rispens) rMDV-EGFP expressing EGFP reporter gene was used as female strain. By homologous recombination, the EGFP reporter gene expression cassette in the viral rMDV-EGFP was replaced by the expression cassette expressing the (ILTV) gB and gD genes of infectious laryngotracheitis virus. Recombinant Marek virus rMDV-ILTgB and rMDV-ILTgD. expressing gB and gD genes were obtained, respectively. By PCR and indirect immunofluorescence assay, it was proved that the target gene inserted in the recombinant virus was correct in size and could express gB and gD target protein. The results of immunological efficacy test showed that the recombinant virus rMDV-ILTgB and rMDV-ILTgD could provide better immune protection against ILTV attack than the attenuated vaccine. Compared with the control group, there were significant differences in the incidence, fatality rate and body weight gain between the chickens immunized with recombinant virus and those in the control group. There were also significant differences in mortality and body weight gain between vaccinated group and routine attenuated vaccine.
