【摘要】：為評估豬繁殖與呼吸綜合征(PRRS)弱毒活疫苗的免疫效果,本研究于規(guī)模豬場選取具有母源抗體的仔豬,接種PRRS弱毒活疫苗后不同時間采集血清,應用RT-PCR檢測PRRS病毒(PRRSV)核酸,應用N-ELISA和G-ELISA兩種試劑盒檢測PRRSV抗體。結果顯示,在免疫接種后60 d內(nèi)可在實驗仔豬血清中檢測到疫苗株核酸,而在整個實驗期間未能檢測到野毒株核酸。仔豬在免疫前具有較高的母源抗體,陽性率達到100%,此后逐漸下降,在免疫后第28 d/60 d(第2次試驗/第1次試驗)降到低點后又穩(wěn)步升高,從免疫后第60 d/90 d到150 d試驗結束時陽性率均達到100%。應用兩種ELISA試劑盒同時檢測329份血清樣品,檢測結果的符合率為96.96%;配對χ~2檢驗,p0.05,兩者差異不顯著;Kappa值為0.81,屬于極強一致性;兩者相關系數(shù)r為0.794,具有顯著線性相關。本研究結果表明,高水平母源抗體豬群接種弱毒活疫苗后取得良好的免疫效果,可選用N-ELISA或G-ELISA試劑盒進行免疫效果評估。
[Abstract]:In order to evaluate the immune effect of porcine reproductive and respiratory syndrome (RRS) (PRRS) attenuated live vaccine, piglets with maternal antibodies were selected from large scale pig farms and sera were collected at different times after inoculation of PRRS attenuated live vaccine. PRRS virus (PRRSV) nucleic acid was detected by RT-PCR and PRRSV antibody was detected by N-ELISA and G-ELISA kits. The results showed that the nucleic acid of the vaccine strain could be detected in the serum of the experimental piglets within 60 days after immunization, but the nucleic acid of the wild strain could not be detected during the whole experiment period. Piglets had high maternal antibody before immunization, the positive rate reached 100, then decreased gradually, and then increased steadily after 28d / 60d (test 2 / test 1). From the 60th day to the 150th day after immunization, the positive rate was 100%. Two kinds of ELISA kits were used to simultaneously detect 329 serum samples. The coincidence rate of the test results was 96.966.The paired 蠂 ~ 2 test showed that there was no significant difference between them, the Kappa value was 0.81which was very consistent. The correlation coefficient r was 0.794, and there was a significant linear correlation between them. The results showed that high level maternal antibody could be used to evaluate the immune response of pigs after inoculation of live attenuated vaccine. N-ELISA or G-ELISA kit could be used to evaluate the immune effect.
