白酒酒糟制備多酶飼料技術(shù)的研究

【摘要】:多酶飼料是生物飼料的一種,飼料中富含多種飼用酶,能夠在一定程度上消除飼料原料中的抗營養(yǎng)成分或?qū)游飪?nèi)源性酶類起到補充作用,從而有利于改善現(xiàn)有飼料的品質(zhì)、提高飼料的利用率。本研究以酒糟為飼料原料、枯草芽孢桿菌和乳酸菌為發(fā)酵菌種、以植酸酶和甘露聚糖酶等為研究對象,通過微生物發(fā)酵的方式制備新型多酶生物飼料。通過對三株枯草芽孢桿菌L1、L2和L3進行單菌單因素和正交固態(tài)發(fā)酵條件優(yōu)化實驗,分別確定了最佳產(chǎn)酶條件:菌株L1單菌固態(tài)發(fā)酵生產(chǎn)甘露聚糖酶的最優(yōu)培養(yǎng)條件為玉米粉和豆粕粉的比例為8:2、酒糟含量為30%、接種量為每瓶0.8mL(250mL裝10g干料)、溫度35℃、pH自然、料水比為1:1.2、培養(yǎng)時間6d;菌株L2單菌固態(tài)發(fā)酵生產(chǎn)木聚糖酶的最優(yōu)培養(yǎng)條件為玉米粉和豆粕粉的比例為6:4、酒糟含量為40%、接種量為每瓶1mL(250mL裝10g干料)、溫度35℃、pH自然、料水比為1:1、培養(yǎng)時間5d;菌株L3單菌固態(tài)發(fā)酵生產(chǎn)植酸酶的最優(yōu)培養(yǎng)條件為:玉米粉和豆粕粉的比例為5:5、酒糟含量為30%、接種量為每瓶0.8mL(250mL裝10g干料)、溫度37℃、pH7、料水比為1:1.2、培養(yǎng)時間5d。根據(jù)單菌發(fā)酵實驗結(jié)果進行混菌發(fā)酵產(chǎn)酶條件的優(yōu)化實驗,確定混菌發(fā)酵最佳產(chǎn)酶條件:玉米粉與豆粕粉比例為7:3、酒糟含量為37%、麩皮含量為30%、接種量為5%、接種方式為三株枯草芽孢桿菌比例為2:2:1,并將植酸酶產(chǎn)生菌提前接種12 h、發(fā)酵溫度37℃、料水比1:1、pH 8.0、添加硫酸銨3%、尿素1.5%、氯化鈣0.3%、培養(yǎng)時間5d 。將經(jīng)過枯草芽孢桿菌發(fā)酵后的飼料進行乳酸菌發(fā)酵培養(yǎng),以改善飼料氣味、和色澤。優(yōu)化后的乳酸菌菌株R1固態(tài)發(fā)酵條件為裝料量100g、溫度35℃、接種量15%、時間6d。培養(yǎng)結(jié)束后,飼料顏色較培養(yǎng)前由黑色變?yōu)榧t棕色,氣味舒適,消除了枯草芽孢桿菌發(fā)酵氣味和氨味。并分別采用DNS法、地衣酚-鹽酸雙波長差值法和TCA法,測定了發(fā)酵前后飼料中甘露聚糖、木聚糖和植酸磷含量的變化情況。發(fā)酵前后甘露聚糖含量平均下降66.28%、木聚糖含量平均下降12.21%、植酸磷含量平均下降34.09%;又對發(fā)酵后的飼料進行粗蛋白、粗纖維、粗灰分、鈣含量、總磷含量、水分等十個指標的測定,測定結(jié)果符合NY/T 1245-2006,即奶牛用精飼料標準。
