九里明超微粉對仔豬生長性能的影響及其機(jī)理初探

【摘要】:本文以九里明為材料,超微粉碎后進(jìn)行粒度檢測、掃描電鏡形態(tài)觀察和總黃酮含量的測定。然后將九里明超微粉添加到仔豬飼料中,研究對仔豬生長性能的影響。選取體重為10 kg左右的杜×長×大三元雜交仔豬90頭,按隨機(jī)區(qū)組設(shè)計、公母各半的原則,將仔豬分成5個處理組,每個處理3個重復(fù),每個重復(fù)6頭仔豬。Ⅰ組飼喂基礎(chǔ)日糧,Ⅱ組飼喂添加75 mg/kg金霉素和20 mg/kg硫酸粘桿菌素的日糧,Ⅲ、Ⅳ、Ⅴ組分別飼喂添加0.4%、0.8%、1.2%九里明超微粉的日糧。進(jìn)行為期30 d的試驗(yàn),研究九里明超微粉對仔豬生長性能、腹瀉頻率、血清生化指標(biāo)、糞便微生物、營養(yǎng)物質(zhì)消化率以及IGF-1 mRNA表達(dá)量的影響。結(jié)果如下:1.九里明超微粉的中心粒徑為8.88μm,90%的超微粉粒徑小于等于23.37μm,符合超微粉碎要求。掃描電鏡觀察發(fā)現(xiàn),超微粉碎處理使九里明植物組織細(xì)胞結(jié)構(gòu)破損顯著,粉體粒度更小,更均勻。2.九里明超微粉總黃酮方法學(xué)試驗(yàn)的相對偏差值良好,適合用此方法檢測九里明超微粉總黃酮。金絲桃苷的標(biāo)準(zhǔn)曲線為y=0.0534x-0.051,在3.2~16μg/mL濃度范圍內(nèi)有良好的線性關(guān)系(R2=0.9975)。試驗(yàn)發(fā)現(xiàn),九里明超微粉總黃酮含量為2.72%。3.添加九里明超微粉能提高仔豬平均日增重和平均日采食量,降低料肉比,添加0.8%的九里明超微粉能顯著提高仔豬的平均日增重、顯著降低料肉比,且與抗生素添加組的效果相當(dāng);添加九里明超微粉降低了仔豬的腹瀉頻率,并且0.8%、1.2%的添加量和添加抗生素降低仔豬腹瀉頻率能達(dá)到顯著水平;添加九里明超微粉能在一定程度上提高血清GLU、TG、T3、T4、IGF-I含量,降低血清BUN含量,其中添加0.8%的九里明超微粉能顯著提高血清T3、IGF-I的含量,顯著降低BUN含量。4.抗生素的添加顯著降低了仔豬糞便中大腸埃希菌數(shù)量,九里明超微粉的添加也不同程度地降低了仔豬糞便中大腸埃希菌數(shù)量,試驗(yàn)30 d時0.8%、1.2%九里明超微粉的添加降低大腸埃希菌數(shù)達(dá)到了顯著水平;與對照組相比,各處理組糞便乳酸菌數(shù)量均有所增加,但差異不顯著;添加不同量的九里明超微粉不同程度地提高了仔豬各營養(yǎng)物質(zhì)的表觀消化率,添加九里明超微粉的各組間相比,添加0.8%的九里明超微粉的效果最好。5.隨著九里明超微粉添加量的增加,仔豬血液IGF-I mRNA相對表達(dá)量也呈增加趨勢;添加九里明超微粉的各試驗(yàn)組血液IGF-I mRNA相對表達(dá)量要高于空白對照組(Ⅰ組),分別提高了18.97%、24.90%、49.80%;與抗生素添加組(Ⅱ組)相比,九里明超微粉添加組血液IGF-I mRNA相對表達(dá)量也有所升高,但未達(dá)到顯著差異的水平。總之,仔豬日糧中添加九里明超微粉能降低仔豬的腹瀉頻率,調(diào)節(jié)了仔豬腸道菌群結(jié)構(gòu),改善了仔豬腸道環(huán)境,增強(qiáng)了仔豬的消化吸收能力,并調(diào)節(jié)IGF-I mRNA的表達(dá),改善了血液生化指標(biāo),促進(jìn)了仔豬的生長性能。結(jié)合九里明超微粉的使用量而言,0.8%的添加量效果最好。九里明超微粉促進(jìn)仔豬生長的可能機(jī)理是:九里明的藥效成分黃酮等化合物抑制了腸道有害菌的增殖,促進(jìn)了有益菌的生長,改善了腸道微生物環(huán)境,進(jìn)而降低仔豬腹瀉頻率、提高了腸道對營養(yǎng)物質(zhì)的消化吸收能力;九里明超微粉作為營養(yǎng)素成分作用于機(jī)體,調(diào)節(jié)IGF-I mRNA的表達(dá),調(diào)節(jié)T3、T4和IGF-I的分泌,影響了機(jī)體對糖類、脂肪和蛋白質(zhì)的代謝。在九里明超微粉藥效和營養(yǎng)的雙重作用下,仔豬的生長性能得到了促進(jìn)。
