轉(zhuǎn)化生長因子β對體外成熟豬卵子成熟率的影響

【摘要】:在豬卵子的體外生產(chǎn)方面,關(guān)于卵子的體外成熟和體外受精技術(shù)目前還存在著不足,特別是豬的受精卵體外生產(chǎn)方面存在復(fù)雜的細(xì)胞質(zhì)成熟、多精子侵入率較高以及前核形成受到抑制等問題。為改善豬卵子體外成熟體系,將轉(zhuǎn)化生長因子β(transforming growth factor-β,簡稱TGFβ)添加到體外成熟的培養(yǎng)液中,研究TGFβ對裸卵和卵丘卵母細(xì)胞復(fù)合體成熟率的影響。在沒有添加TGFβ的體外成熟培養(yǎng)液中,裸卵和卵丘卵母細(xì)胞復(fù)合體被成熟培養(yǎng)24 h,結(jié)果沒有卵子達到M-Ⅱ時期,但是在添加TGFβ的體外成熟培養(yǎng)液中,觀察到有卵子達到M-Ⅱ時期。另一方面,當(dāng)卵丘卵母細(xì)胞復(fù)合體在體外成熟培養(yǎng)時,將TGFβ添加到不同培養(yǎng)階段(0~24 h或24~48 h)的培養(yǎng)液時,卵子的成熟率沒有差別。對裸卵在體外成熟培養(yǎng)24 h時,當(dāng)在前半期添加TGFβ時(0~24 h),卵子的成熟率為59%;當(dāng)在后半期添加TGFβ時(24~48 h),卵子的成熟率為57%。同樣在裸卵組中,當(dāng)在全期添加TGFβ時(0~48 h),卵子的成熟率為27%;當(dāng)無添加TGFβ時,卵子的成熟率為38%。前2組與后2組相比,卵子成熟率有顯著差異。
