硫酸安普霉素可溶性粉中非法添加乙酰甲喹的HPLC-PDA檢測方法的建立

【摘要】:為檢測硫酸安普霉素可溶性粉中非法添加的乙酰甲喹,以十八烷基硅烷鍵合硅膠為填充劑,磷酸鹽緩沖液(取磷酸二氫鈉3.0 g,加水1000 m L使溶解,加三乙胺0.5 m L,用飽和氫氧化鈉溶液調(diào)節(jié)p H值至7.0)-甲醇為流動(dòng)相,二極管陣列檢測器(PDA),建立了HPLC-PDA檢測方法,并采用峰純度檢查和光譜相似度檢查輔助對照品比對方法,對非法添加藥物進(jìn)行確證。在此液相色譜條件下,乙酰甲喹與其他物質(zhì)峰分離良好。按外標(biāo)法以峰面積計(jì)算,乙酰甲喹的平均回收率為98.1%,RSD為0.3%。該檢測方法簡便、準(zhǔn)確、可靠,可用于測定硫酸安普霉素可溶性粉中非法添加的乙酰甲喹。
[Abstract]:In order to detect the illegal addition of acetylmethaquine in the soluble powder of ampoumycin sulfate, using octadecyl silane bonded silica gel as filler, phosphate buffer solution (sodium dihydrogen phosphate 3.0 g, water 1000 mL) was added to dissolve, and triethylamine was added to 0.5 mL. Using saturated sodium hydroxide solution to adjust pH to 7.0) -methanol as mobile phase and diode array detector (PDA),) as mobile phase, a HPLC-PDA detection method was established. Confirm the illegal addition of drugs. Under this liquid chromatographic condition, the peak of acetylmethaquine was well separated from other substances. According to the external standard method, the average recovery of acetomequine was 98.1% and RSD was 0.3%. The method is simple, accurate and reliable. It can be used for the determination of acetomequine in the soluble powder of ampouamycin sulfate.
