【摘要】：實(shí)驗(yàn)旨在建立快速檢測(cè)驢、馬和狐貍源性成分的方法。以驢、馬和狐貍的線(xiàn)粒體保守序列16S r RNA基因?yàn)榘谢?設(shè)計(jì)通用引物和以不同熒光素標(biāo)記的特異Taq Man探針,構(gòu)建與通用引物共用的陽(yáng)性擴(kuò)增內(nèi)標(biāo)(Internal Amplification Control,IAC)作為PCR監(jiān)控反應(yīng)體系。通過(guò)優(yōu)化PCR反應(yīng)體系和條件,建立能同時(shí)檢測(cè)驢、馬和狐貍源性成分的四重實(shí)時(shí)熒光PCR方法。結(jié)果表明:利用驢、馬、狐貍、牛、豬、羊、水貂、狗、雞、鴨、大豆、小麥、玉米等物種檢測(cè)沒(méi)有交叉反應(yīng),證明該方法特異性高,準(zhǔn)確性好。該方法模板DNA檢測(cè)靈敏度為0.01 ng。本研究建立的驢、馬和狐貍源性四重?zé)晒鈱?shí)時(shí)PCR檢測(cè)體系靈敏度高、特異性強(qiáng),能有效鑒別動(dòng)物產(chǎn)品中驢、馬和狐貍的源性成分,也可用作相關(guān)動(dòng)物皮張真?zhèn)蔚蔫b別方法。
[Abstract]:The aim of this study was to establish a rapid method for the detection of donkeys, horses and foxes. Using the mitochondrial conserved 16s r RNA gene of donkey, horse and fox as target gene, a universal primer and a specific Taq Man probe labeled with different fluorescein were designed to construct a positive amplified (Internal Amplification Control, shared with universal primers. IAC) as PCR monitoring reaction system. By optimizing the PCR reaction system and conditions, a quadruple real-time fluorescent PCR method was established for simultaneous detection of donkey, horse and fox origin components. The results showed that there was no cross reaction among donkey, horse, fox, cow, pig, sheep, mink, dog, chicken, duck, soybean, wheat and corn. The sensitivity of template DNA is 0. 01 ng.. The real-time PCR system for the detection of donkey, horse and fox is highly sensitive and specific. It can be used to identify the origin components of donkey, horse and fox in animal products, and can also be used as a method to identify the authenticity and falsity of related animal skin.
