模擬運輸應激對大鼠肺臟免疫機能及TLR4-NF-κB信號通路的影響

【摘要】:現(xiàn)代養(yǎng)殖的集約化程度不斷提高,導致動物的長途運輸變得越來越普遍。由于長途運輸途中禁食或限飼、噪音、高溫、顛簸、心理壓力等因素的綜合作用,常引起動物產(chǎn)生嚴重的運輸應激反應,降低了動物的生產(chǎn)性能、免疫機能和肉品質量,造成重大經(jīng)濟損失以及健康隱患。因此,本研究選用大鼠作為實驗動物,制作大鼠模擬運輸應激模型,以研究運輸應激對動物的肺臟組織及肺泡巨噬細胞免疫功能的影響。1.將SD大鼠適應飼養(yǎng)7d后,于每日9:00-11:00將模擬運輸組大鼠置于35℃、60r/min的水平搖床振蕩搖晃,連續(xù)3天,模擬運輸過程中的搖晃、高溫、噪音、擁擠等因素,構建大鼠運輸模型,以觀察模擬運輸應激對大鼠的臨床癥狀、體重、血清TNF-α、IL-6、皮質酮及肺臟組織結構的影響。結果表明:(1)模擬運輸應激可引起大鼠精神緊張,行動遲緩,全身發(fā)抖,被毛凌亂,呼吸急促,尿量減少且發(fā)黃,糞便干結且萎縮。(2)模擬運輸應激可引起大鼠體重顯著低于對照組(P0.05)。(3)模擬運輸應激可使大鼠血清TNF-α和 IL-6濃度升高,且IL-6顯著高于對照組(P0.05)。(4)模擬運輸應激可引起大鼠血清皮質酮濃度顯著高于對照組(P0.05)。(5)模擬運輸應激可造成大鼠肺泡不均勻擴張,部分肺泡間隔斷裂;小靜脈擴張充血,炎癥細胞浸潤,部分肺泡腔內可見水腫。因此,經(jīng)每日35℃、60r/min搖晃兩個小時,連續(xù)3天,可引起大鼠出現(xiàn)嚴重的運輸應激及肺組織損傷。2.通過提取大鼠肺臟組織mRNA,經(jīng)反轉錄擴增后,進行相關免疫指標基因擴增,檢測IL-1β、IL-6、TNF-α、IFN-γ、TLR4、β-actin的1nRNA表達量,結果表明:模擬運輸應激能顯著降低肺臟組織TNF-α、IFN-γ mRNA的表達量(P0.05);同時TLR4、IL-1β、IL-6 mRNA的表達量略有增加,但與對照組相比并不顯著,說明模擬運輸應激可抑制大鼠肺臟組織的免疫功能。3.大鼠模擬運輸應激造模結束后,采用反復沖洗法收集大鼠肺泡巨噬細胞,抽提大鼠肺泡巨噬細胞總蛋白,Western Blot法檢測NF-κB p65、Phospho-NF-κB p65、IκBα、Actin蛋白表達量。結果表明:模擬運輸應激可提高肺泡巨噬細胞NF-κB p65磷酸化水平,顯著降低抑制蛋白IκBα的表達量(P0.05),說明模擬運輸應激可激活肺泡巨噬細胞的NF-κB信號通路,將誘導下游前炎性細胞因子的表達和分泌。因此,模擬運輸應激可引起大鼠血液皮質酮含量急劇升高,損傷肺臟組織結構,并抑制肺臟免疫機能,可激活肺泡巨噬細胞NF-κB信號通路,誘導前炎性細胞因子的分泌和表達。
