【摘要】：為探討抗生素造模對菌群失調(diào)小鼠腸道乳糖酶活性的影響,本研究采用硫酸慶大霉素和頭孢拉定混合抗生素制備菌群失調(diào)腹瀉小鼠模型,造模成功后分別提取模型組和正常組小鼠第1 d至第7 d腸道內(nèi)容物及腸粘膜。將腸粘膜平均分成前、中、后3段,每段約10 cm,用ONPG法分別測定腸內(nèi)容物及各段腸黏膜所含乳糖酶活性,結(jié)果表明,造模第2 d,模型組腸內(nèi)容物中乳糖酶活性均明顯低于正常組,第4 d開始兩組差異顯著(p0.05)。模型組腸前段黏膜乳糖酶活性自第3 d開始與正常組對比差異極顯著(p0.01)、腸中段、腸后段黏膜乳糖酶活性分別自第2 d、第3 d開始與正常組對比差異顯著(p0.05),且各腸段粘膜自第5 d開始與正常組對比差異極顯著(p0.01)。上述結(jié)果表明,抗生素造模顯著影響小鼠腸道乳糖酶活性,且抗生素所致菌群失調(diào)性腹瀉與腸道乳糖酶活性降低有關(guān)。
[Abstract]:In order to investigate the effect of antibiotic modeling on lactase activity in intestinal tract of mice with dysbacteriosis, the model of diarrhea was established by using mixed antibiotics of gentamicin sulfate and cefradine. The intestinal contents and intestinal mucosa were extracted from the model group and the normal group from day 1 to day 7. The activity of lactase in intestinal contents and intestinal mucosa was measured by ONPG method for 10 cm, the results showed that the activity of lactase in intestinal mucosa was determined at the 2nd day of model making, and the results showed that the activity of lactase in intestinal mucosa was determined by ONPG method. The lactase activity in the intestinal contents of the model group was significantly lower than that in the normal group, and there was a significant difference between the two groups at the 4th day (p0.05). The activity of lactase in the anterior mucosa of the model group was significantly different from that in the normal group from the 3rd day (p0.01), and the activity of lactase in the middle and posterior segments of the intestine was from the 2nd day, respectively. There was a significant difference between the third day and the normal group (p0.05), and the significant difference between the intestinal mucosa and the normal group from the 5th day (p0.01). The results indicated that antibiotic modeling significantly affected the intestinal lactase activity in mice, and the decrease of intestinal lactase activity was associated with the decrease of intestinal lactase activity in patients with dysbacteriosis induced by antibiotics.
