【摘要】：為建立能同時(shí)檢測(cè)犬圓環(huán)病毒(CanineCV)和犬細(xì)小病毒(CPV)的方法,根據(jù)CanineCV和CPV基因組的保守區(qū)域設(shè)計(jì)2對(duì)特異性引物,預(yù)期特異性擴(kuò)增CanineCV和CPV的片段大小分別為984和668 bp。并將反應(yīng)條件進(jìn)行優(yōu)化,建立了CanineCV和CPV的雙重PCR檢測(cè)方法。特異性檢測(cè)結(jié)果顯示,該方法能特異性擴(kuò)增CanineCV和CPV的目的條帶,且未擴(kuò)增出其他犬胃腸道病原;CanineCV和CPV的最低檢出限分別為85.7和312 copies/L。對(duì)采集自廣西地區(qū)的223份臨床樣品的檢測(cè)結(jié)果顯示,CanineCV的陽性率為2.24%(5/223)、CPV的陽性率為5.38%(12/223),后者與常規(guī)PCR檢測(cè)方法相一致。結(jié)果表明,建立的雙重PCR方法可以用于CanineCV和CPV的檢測(cè)和流行病學(xué)調(diào)查。
[Abstract]:In order to establish a method for simultaneous detection of canine circovirus (CanineCV) and canine parvovirus (CPV), two pairs of specific primers were designed according to the conserved region of CanineCV and CPV genomes. The expected length of CanineCV and CPV fragments were 984 and 668 bp., respectively. The reaction conditions were optimized and a dual PCR detection method for CanineCV and CPV was established. The results of specific detection showed that the target bands of CanineCV and CPV could be amplified by this method, and the detection limits of CanineCV and CPV were 85.7 and 312 copies/L., respectively. The positive rate of CanineCV was 2.24% (5 / 223), CPV, 5.38%, 12 / 223), which was consistent with the conventional PCR method. The results showed that the dual PCR method could be used for the detection and epidemiological investigation of CanineCV and CPV.
