轉(zhuǎn)Bt基因水稻秸稈對(duì)小鼠代謝、發(fā)育和繁殖的安全性評(píng)價(jià)

【摘要】:本研究利用轉(zhuǎn)Bt基因水稻秸稈(秸稈粉與小鼠普通飼料成分按質(zhì)量比1:3混合配制成飼料)喂養(yǎng)小鼠90d,通過(guò)對(duì)小鼠的代謝、發(fā)育與繁殖等方面的分析,確定Bt蛋白對(duì)哺乳動(dòng)物小鼠的影響,探討轉(zhuǎn)Bt基因水稻秸稈應(yīng)用于飼料生產(chǎn)的安全性問(wèn)題。1.所有小鼠在90天的飼養(yǎng)過(guò)程中飲食、飲水和日常行為均表現(xiàn)正常,沒(méi)有發(fā)生中毒或死亡現(xiàn)象;2.血[相、血生化和血液流變學(xué)等相關(guān)指標(biāo),90d小鼠試驗(yàn)組和對(duì)照組差異均不顯著;3.利用ELISA酶聯(lián)免疫技術(shù)測(cè)出90d小鼠血清中CAT、SOD、POD、GSH-Px和AchE酶活,結(jié)果發(fā)現(xiàn)試驗(yàn)組和對(duì)照組差異均不顯著;4.利用流式細(xì)胞術(shù)(FCM)檢測(cè)90d小鼠血淋巴細(xì)胞的凋亡與鈣流情況,結(jié)果發(fā)現(xiàn)無(wú)論是細(xì)胞凋亡還是鈣離子流失試驗(yàn)組與對(duì)照組差異均不顯著;5.將90d小鼠解剖取主要臟器,觀察臟器病變、檢測(cè)臟器指數(shù),同時(shí)利用ELISA酶聯(lián)免疫技術(shù)測(cè)定各臟器中CAT、SOD、POD、GSH-Px和AchE酶活,結(jié)果發(fā)現(xiàn)臟器無(wú)病害、毒害情況,各臟器指數(shù)和CAT、SOD、POD、GSH-Px、AchE酶活試驗(yàn)組與對(duì)照組均無(wú)顯著性差異;6.取90d雄鼠的精液,測(cè)定其精子密度、活力、活率、畸形率和頂體完整率,結(jié)果發(fā)現(xiàn)試驗(yàn)組與對(duì)照組均無(wú)明顯影響;7.取90d雌鼠的卵巢制作石蠟切片,觀察組織病變情況并統(tǒng)計(jì)原始卵泡、初級(jí)卵泡、次級(jí)卵泡和三級(jí)卵泡數(shù)量,結(jié)果發(fā)現(xiàn)試驗(yàn)組與對(duì)照組卵巢組織沒(méi)有明顯病變情況,各級(jí)卵泡數(shù)量豐富且無(wú)顯著性差異。研究表明,轉(zhuǎn)Bt基因水稻秸稈對(duì)小鼠沒(méi)有產(chǎn)生毒害作用,沒(méi)有引起小鼠各臟器的病變,也沒(méi)有刺激小鼠體內(nèi)的關(guān)鍵代謝酶的解毒機(jī)制從而引起幾種主要酶酶活的變化,并且對(duì)于雌性和雄性生殖健康也沒(méi)有明顯有害影響。
[Abstract]:In this study, Bt transgenic rice straw was used to feed mice for 90 days, and the metabolism, development and reproduction of mice were analyzed. To determine the effect of Bt protein on mammalian mice and to explore the safety of transgenic rice straw with Bt gene in feed production. 1. All the mice showed normal diet, drinking water and daily behavior during the 90-day feeding, and no poisoning or death occurred. 2. There was no significant difference in blood phase, blood biochemistry and hemorheology between the test group and the control group. The activities of CAT,SOD,POD,GSH-Px and AchE in serum of 90 day mice were measured by ELISA enzyme-linked immunosorbent assay. The results showed that there was no significant difference between the test group and the control group. 4. Flow cytometry (FCM) was used to detect the apoptosis and calcium flow in blood lymphocytes of 90 day mice. The results showed that there was no significant difference between the experimental group and the control group in both apoptosis and calcium ion loss. 5. The main organs of mice were dissected for 90 days, the pathological changes of organs were observed, and the index of organs was detected. Meanwhile, the activities of CAT,SOD,POD,GSH-Px and AchE in various organs were determined by ELISA enzyme-linked immunosorbent assay. The results showed that there was no disease and toxicity in the organs. There was no significant difference in organ index and CAT,SOD,POD,GSH-Px,AchE enzyme activity between the test group and the control group. 6. The sperm density, motility, viability, deformity rate and acrosomal integrity rate were measured in 90 day male rats. The results showed that there was no significant effect on sperm density, motility, acrosome integrity, and no significant difference between the experimental group and the control group. Paraffin sections were made from the ovaries of 90 d female rats. The pathological changes of the tissues were observed and the number of primary follicles, secondary follicles and tertiary follicles were counted. The results showed that there was no obvious pathological changes in the ovarian tissues between the experimental group and the control group. There was no significant difference in the number of follicles at all levels. The results showed that rice straw transformed with Bt gene had no toxic effect on mice, did not cause changes in various organs of mice, nor did it stimulate the detoxification mechanism of key metabolic enzymes in mice, thus causing changes in the activities of several major enzymes. There were also no significant adverse effects on reproductive health of both females and males.
