神經(jīng)介素U在體外對(duì)豬淋巴細(xì)胞活性及自然殺傷細(xì)胞活性的影響

【摘要】:[目的]本文旨在研究神經(jīng)介素U(NMU)對(duì)豬外周血淋巴細(xì)胞(PBL)的免疫調(diào)節(jié)作用。[方法]以小梅山豬為研究對(duì)象,用半定量RT-PCR方法研究了豬PBL中NMU及其受體(NMUR1和NMUR~2)基因的表達(dá),用免疫組織化學(xué)染色方法檢測(cè)了豬PBL中NMU及其受體蛋白的分布,用Cell Counting Kit-8(CCK-8)細(xì)胞計(jì)數(shù)法研究了NMU對(duì)豬PBL細(xì)胞和NK細(xì)胞活性的影響。[結(jié)果]NMUR1 mRNA在豬PBL中表達(dá),NMU和NMUR1蛋白在豬PBL的胞膜及胞漿均有分布。用不同濃度(0.01、0.1、1、10、100和1 000 nmol·L~(-1))NMU處理體外培養(yǎng)的PBL,與對(duì)照組相比,0.1、1和10 nmol·L~(-1)NMU極顯著提高豬PBL的細(xì)胞活性(P0.01),且當(dāng)NMU濃度為10 nmol·L~(-1)時(shí),PBL活性的水平達(dá)到峰值(179.4±3.0)%。用不同濃度(0.01、0.1、1、10、100和1 000 nmol·L~(-1))NMU處理體外培養(yǎng)的PBL細(xì)胞作為效應(yīng)細(xì)胞,以K562細(xì)胞作為靶細(xì)胞。與對(duì)照組相比,在效靶比為15∶1時(shí),0.1、1和10 nmol·L~(-1)的NMU均能顯著提高NK細(xì)胞的細(xì)胞活性(P0.01);效靶比為30∶1時(shí),1和10 nmol·L~(-1)的NMU均能顯著提高NK細(xì)胞的細(xì)胞活性(P0.01);在效靶比為60∶1時(shí),10 nmol·L~(-1)NMU能顯著提高NK細(xì)胞的細(xì)胞活性(P0.01)。[結(jié)論]NMU及NMUR1在豬PBL中均有表達(dá),且NMU在一定濃度范圍內(nèi)能提高豬NK細(xì)胞對(duì)K562細(xì)胞的殺傷能力。
[Abstract]:[objective] to study the immunomodulatory effect of interleukin U (NMU) on (PBL) in porcine peripheral blood lymphocytes. [methods] the expression of NMU and its receptor (NMUR1 and NMUR~2) genes in porcine PBL was studied by semi-quantitative RT-PCR method, and the distribution of NMU and its receptor protein in PBL was detected by immunohistochemical staining. The effects of NMU on the activity of porcine PBL and NK cells were studied by Cell Counting Kit-8 (CCK-8) cell count method. [results] NMUR1 mRNA was expressed in porcine PBL and NMU and NMUR1 proteins were distributed in the membrane and cytoplasm of porcine PBL. Compared with the control group, the cell activity of porcine PBL was significantly increased by 0.1 nmol L-1 NMU and 0.1 nmol L-1 NMU, and the PBL activity reached a peak value of (179.4 鹵3.0)% when the NMU concentration was 10 nmol L-1 at different concentrations (0.01 nmol L ~ (-1) and 1 000 nmol L ~ (-1) NMU). The PBL cells cultured in vitro were treated with different concentrations (0. 01 nmol L ~ (-1) NMU) and 1 000 nmol L ~ (-1) NMU as effector cells and K562 cells as target cells. Compared with the control group, At 15:1, NMU with 0.1 and 10 nmol L-1 significantly increased the cellular activity of NK cells (P0.01), NMU with 1 and 10 nmol L-1 significantly increased the cellular activity of NK cells at 30:1, and 10 nmol L-1 NMU at 60:1. The cell activity of NK cells was significantly increased (P0.01). [conclusion] both NMU and NMUR1 are expressed in porcine PBL, and NMU can enhance the cytotoxicity of porcine NK cells to K562 cells in a certain concentration range.
