金絲桃苷對小鼠T淋巴細(xì)胞亞群及血清細(xì)胞因子的影響

【摘要】:觀察金絲桃苷對小鼠免疫系統(tǒng)的作用,將Balb/c小鼠隨機(jī)分成5組,空白對照組(在第1、3、5天腹腔注射生理鹽水)、免疫抑制組(在第1、3、5天腹腔注射環(huán)磷酰胺)、給藥組(在第1、3、5天腹腔注射環(huán)磷酰胺,第1~7天每天以150/300mg/kg金絲桃苷水溶液灌胃)和陽性對照組(在第1、3、5天腹腔注射環(huán)磷酰胺,第1~7天每天以0.2mg/kg黃芪多糖水溶液灌胃)。用ELISA法測定小鼠T淋巴細(xì)胞亞群CD_3、CD_4、CD_8以及細(xì)胞因子IL-4、IL-6、IL-10、IFN-γ、TNF-α的水平。結(jié)果表明,與空白對照組相比,免疫抑制組的CD_3、CD_4、CD_8水平以及血清細(xì)胞因子IFN-γ、TNF-α水平顯著降低(P0.05),IL-4、IL-6及IL-10水平顯著升高(P0.05)。給予金絲桃苷后第8天,與免疫抑制組相比,CD_3、CD_4、CD_8水平以及血清細(xì)胞因子IFN-γ、TNF-α水平極顯著升高(P0.01),IL-4、IL-6、IL-10水平極顯著降低(P0.01)。金絲桃苷能通過提高免疫抑制小鼠T淋巴細(xì)胞亞群的CD_3、CD_4、CD_8水平以及細(xì)胞因子IFN-γ及TNF-α水平,降低IL-4、IL-6、IL-10水平,來拮抗環(huán)磷酰胺所致的免疫機(jī)能抑制,提高機(jī)體免疫功能。
[Abstract]:To observe the effect of hyperin on the immune system of mice, Balb/c mice were randomly divided into 5 groups. The blank control group (intraperitoneal injection of normal saline on the 1st day 3 days), the immunosuppressive group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), the administration group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), the immunosuppressive group (intraperitoneal injection of cyclophosphamide on the 1st day 3 days), 150/300mg/kg hyperoside solution was administered daily on day 1 to day 7, and the positive control group was given cyclophosphamide intraperitoneally on day 1 and day 7 respectively. 0.2mg/kg Astragalus membranaceus polysaccharide solution was administered intragastrically on day 1 and day 7 respectively. The levels of CD_3,CD_4,CD_8 and cytokines IL-4,IL-6,IL-10,IFN- 緯 and TNF- 偽 in T lymphocyte subsets of mice were determined by ELISA method. The results showed that compared with the control group, the levels of CD_3,CD_4,CD_8 and serum cytokines IFN- 緯 and TNF- 偽 in immunosuppressive group were significantly decreased (P0.05), and the levels of IL-4,IL-6 and IL-10 were significantly increased (P0.05). On the 8th day after hypericin administration, the levels of CD_3,CD_4,CD_8, serum cytokines IFN- 緯 and TNF- 偽 were significantly increased (P0.01) and IL-4,IL-6,IL-10 levels were significantly decreased (P0.01) compared with the immunosuppressive group. Hypericin can antagonize the immunosuppression of cyclophosphamide by increasing the level of CD_3,CD_4,CD_8 of T lymphocyte subsets, the levels of cytokines IFN- 緯 and TNF- 偽, and decreasing the level of IL-4,IL-6,IL-10, which can antagonize the inhibition of immune function induced by cyclophosphamide and improve the immune function of the body.
