益生菌增加IPEC-J2細胞表面半乳糖在抑制RV感染中的作用

【摘要】:輪狀病毒通過識別細胞表面糖鏈附著于腸上皮細胞,一些腸道內(nèi)常駐菌群可以修改細胞表面糖鏈。以3種益生菌上清孵育豬小腸上皮細胞IPEC-J2,豬輪狀病毒(PRV)感染細胞。采用凝集素熒光技術(shù)測定各處理組IPEC-J2細胞表面半乳糖,檢測對應(yīng)輪狀病毒滴度和RV含量變化。結(jié)果表明,對照組PRV TCID_(50)·0.1 mL~(-1)為10~(-7.15±0.14),干酪乳桿菌組TCID_(50)·0.1mL~(-1)為10~(-3.875±0.125),食淀粉乳桿菌PRV TCID_(50)·0.1 mL~(-1)為10~(-4.125±0.138),枯草芽孢桿菌PRV TCID_(50)·0.1 mL~(-1)為10~(-4.16±0.144)。3組益生菌上清組在RV感染48 h內(nèi),RV量顯著低于未處理組。凝集素熒光標記表明,3株益生菌上清均可改變細胞表面半乳糖含量。干酪乳桿菌、食淀粉乳桿菌和枯草芽孢桿菌上清可增加IPEC-J2細胞表面半乳糖含量,提高細胞抗RV感染能力,阻斷RV黏附,保護腸道上皮細胞。
[Abstract]:Rotavirus adheres to intestinal epithelial cells by recognizing the sugar chains on the surface of the cells, and some intestinal resident bacteria can modify the sugar chains on the surface of the cells. Porcine intestinal epithelial cells (IPEC-J2,) infected with porcine rotavirus (PRV) were incubated with three probiotic supernatants.
