【摘要】：本試驗采用超濾法結(jié)合高效液相色譜-串聯(lián)質(zhì)譜法(HPLC-MS/MS)測定沃尼妙林與豬、雞和兔血漿蛋白結(jié)合率,并對血漿中不同濃度沃尼妙林的蛋白結(jié)合率進行比較。結(jié)果表明,建立的液相色譜-串聯(lián)質(zhì)譜聯(lián)用法用于沃尼妙林生物樣品的測定具有簡便、靈敏與選擇性高的特點。沃尼妙林具有較強的血漿蛋白結(jié)合率,其在豬、雞和兔血漿中的平均蛋白結(jié)合率分別為84.4%±1.07%,87.5%±1.83%和86.6%±1.96%。沃尼妙林的血漿蛋白結(jié)合率在0.1~10μg/m L的考察濃度范圍內(nèi)無顯著性差異,蛋白結(jié)合呈現(xiàn)非濃度依賴的特性。
[Abstract]:In this study, the plasma protein binding rates of Vornimarin to pigs, chickens and rabbits were determined by ultrafiltration and high performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS), and the protein binding rates of different concentrations were compared. The results showed that the method was simple, sensitive and selective. The average protein binding rate in pig, chicken and rabbit plasma was 84.4% 鹵1.07% and 87.5% 鹵1.83% and 86.6% 鹵1.96%, respectively. There was no significant difference in the plasma protein binding rate in the concentration range of 0.1 ~ 10 渭 g / mL, and the protein binding was non-concentration-dependent.
