【摘要】：為建立快速、準(zhǔn)確、簡(jiǎn)便的檢測(cè)氣腫疽的膠體金免疫層析試紙條,以免疫層析法為指導(dǎo),根據(jù)抗原-抗體的特性,建立了不需要再加入任何檢測(cè)試劑且快速簡(jiǎn)便的檢測(cè)方法。結(jié)果表明,膠體金與該單抗的最佳結(jié)合量為30μg/m L,最佳pH值為8.4,最佳NC膜為Millipore HFB1350220型。將純化的氣腫疽梭菌多抗及羊抗鼠IgG分別以1∶4和1∶8的稀釋倍數(shù)噴于硝酸纖維素膜的T線和C線。對(duì)膠體金診斷試紙條進(jìn)行驗(yàn)證,敏感度高,可檢測(cè)到的最低抗原量為1×10~5CFU/m L,特異性強(qiáng)、無假陽性與交叉反應(yīng)現(xiàn)象、有較好的穩(wěn)定性。本研究旨在建立快速、準(zhǔn)確、簡(jiǎn)便的檢測(cè)氣腫疽的膠體金免疫層析試紙條。
[Abstract]:In order to establish a rapid, accurate and simple gold immunochromatographic strip for the detection of emphysema, a rapid and simple method was established based on the characteristics of antigen-antibody and immunochromatographic method. The results show that the best binding amount of colloidal gold to the McAb is 30 渭 g / mL, the optimum pH value is 8.4, and the best NC film is Millipore HFB1350220 type. Purified Clostridium emphysema polyclonal antibody and sheep anti mouse IgG were sprayed on the T line and C line of cellulose nitrate membrane at 1:4 and 1:8 dilution times, respectively. The test strip of colloidal gold diagnosis was proved to be sensitive, the lowest antigen was 1 脳 10~5CFU/m L, the specificity was strong, there was no false positive and cross reaction phenomenon, and there was good stability. The aim of this study was to establish a rapid, accurate and simple gold immunochromatographic strip for the detection of emphysema.
