丁香葉總酚酸提取工藝的優(yōu)化及抗氧化活性測(cè)定

【摘要】:丁香葉主要來(lái)源于木樨科丁香屬植物紫丁香(Syringa oblate Lindl)、朝鮮丁香(Syringa diatata Nakai)和洋丁香(Syringa vulgaris L)的干燥葉。丁香葉具有良好的抗菌消炎、抗氧化、抗病毒、保肝利膽、增強(qiáng)機(jī)體體液免疫、降溫降壓和鎮(zhèn)咳祛痰等作用,是一種理想的廣譜抗菌藥,其藥效已得到肯定。丁香葉膠囊劑和片劑已收錄于《中華人民共和國(guó)衛(wèi)生部藥品標(biāo)準(zhǔn)》中藥成方制劑,總酚酸作為發(fā)揮作用的藥效基礎(chǔ),由于具有較強(qiáng)的抗氧化活性,已在食品領(lǐng)域中廣泛應(yīng)用。但丁香葉中總酚酸含量較低,國(guó)內(nèi)外目前均無(wú)關(guān)于丁香葉總酚酸富集工藝的研究報(bào)道,因而限制了丁香葉開發(fā)和利用。因此,本實(shí)驗(yàn)對(duì)丁香葉提取、富集工藝進(jìn)行優(yōu)化,并驗(yàn)證總酚酸富集后抗氧化活性得到了顯著提高。為丁香葉的工業(yè)化生產(chǎn)及作為新型的保健品和食品添加劑的開發(fā)使用提供理論基礎(chǔ)。本試驗(yàn)首先采用Box Behnken響應(yīng)面法對(duì)丁香葉總酚酸的乙醇提取工藝進(jìn)行優(yōu)化。以總酚酸提取率為考察指標(biāo),在單因素實(shí)驗(yàn)基礎(chǔ)上,采用響應(yīng)面分析法對(duì)乙醇濃度、提取溫度、提取時(shí)間和液料比等提取條件進(jìn)行了優(yōu)化。結(jié)果表明,乙醇回流提取總酚酸的最佳工藝條件為:乙醇濃度44%、提取溫度90℃、提取時(shí)間96min、液料比25.2:1(m L/g)、提取次數(shù)為2次,在此條件下提取率為1.64±0.23mg/g,與預(yù)測(cè)值相近。在提取工藝優(yōu)化的基礎(chǔ)上,本試驗(yàn)采用大孔樹脂法對(duì)丁香葉總酚酸的富集工藝進(jìn)行了優(yōu)化。選取了D-101、X-5、HPD100、HPD 300、AB-8、S-8、NKA-9、HPD826、HPD130、HPD400和HPD600十一種樹脂考察吸附特性。靜態(tài)吸附試驗(yàn)表明S-8樹脂表現(xiàn)出了良好的吸附、解吸能力和快速的吸附能力。用動(dòng)態(tài)吸附、解吸試驗(yàn)優(yōu)化富集工藝,優(yōu)化的吸附條件:總酚酸初始濃度為2.06mg/m L;p H5.0;上樣量4.3BV;上樣速率2BV/h;優(yōu)化的解吸參數(shù)為:去離子水、5BV、50%乙醇,流速2BV/h。按照優(yōu)化的條件用S-8樹脂處理過(guò)的丁香葉總酚酸干燥提取物中含量由2.75%上升到了9.18%,回收率為81.61%。試驗(yàn)結(jié)果表明S-8樹脂對(duì)丁香葉總酚酸具有很好的富集能力,建立的方法穩(wěn)定、可靠,為規(guī);a(chǎn)提供了基礎(chǔ)。使用DPPH·清除率、羥自由基清除率、超氧陰離子自由基清除率、Fe還原能力為考察指標(biāo)來(lái)測(cè)定丁香葉總酚酸純化前后的抗氧化活性。結(jié)果表明丁香葉總酚酸濃度越高,抗氧化活性越好,并且相同質(zhì)量濃度的提取物,純化后的抗氧化活性顯著高于純化前的活性,丁香葉具有較強(qiáng)的抗氧化能力。因此丁香葉可作為一種新型天然抗氧化劑。本研究首次采用響應(yīng)面法優(yōu)化了丁香葉總酚酸的提取工藝,并用大孔吸附樹脂法優(yōu)化了總酚酸的富集工藝,并測(cè)定了其抗氧化活性。為丁香葉總酚酸未來(lái)的產(chǎn)業(yè)化應(yīng)用奠定理論基礎(chǔ)。
[Abstract]:The clove leaves are mainly derived from the dried leaves of (Syringa oblate Lindl), (Syringa diatata Nakai) and (Syringa vulgaris L). Clove leaf is an ideal broad-spectrum antimicrobial drug with good anti-inflammatory, anti-oxidation, anti-virus, anti-liver and gallbladder, enhanced humoral immunity, lower temperature and lower blood pressure, and antitussive and expectorant effect. Clove leaf capsules and tablets have been included in the traditional Chinese medicine prescription of the Ministry of Health of the people's Republic of China. The total phenolic acid has been widely used in the field of food because of its strong antioxidant activity. The content of total phenolic acid in the leaves of Dante is relatively low, which is not related to the enrichment process of total phenolic acid in clove leaves at home and abroad, which limits the development and utilization of clove leaves. Therefore, the extraction and enrichment process of clove leaves was optimized, and the antioxidant activity of total phenolic acid was significantly improved. It provides a theoretical basis for the industrial production of clove leaves and the development and application of new health products and food additives. In this experiment, Box Behnken response surface method was used to optimize the ethanol extraction process of total phenolic acid in clove leaves.
