【摘要】：根據(jù)GenBank中已公布的豬德爾塔冠狀病毒(PDCoV)N基因序列設(shè)計并合成特異的引物和TaqMan探針,構(gòu)建質(zhì)粒標準品,以標準品為模板建立了檢測PDCoV的TaqMan實時熒光定量PCR方法,并對該方法的敏感性、特異性及重復性進行了評價。結(jié)果顯示,以豬流行性腹瀉病毒、豬傳染性胃腸炎病毒、豬庫布病毒、豬繁殖與呼吸綜合征病毒和口蹄疫病毒為模板時均未檢測到熒光信號,表明該方法具有良好的特異性;用2.66×10~6、2.66×10~5和2.66×10~4copies/L這3個不同濃度的質(zhì)粒標準品進重復試驗,循環(huán)閾值Ct的變異系數(shù)均低于2%,表明該方法具有良好的特異性;標準曲線斜率為-3.461,相關(guān)系數(shù)為R~2=0.998,表明閾值和模板濃度之間具有良好的線性關(guān)系。10倍梯度稀釋質(zhì)粒標準品,檢測的最低限度為2.66×10~1 copies/L的質(zhì)粒DNA,表明此方法具有良好的敏感性。利用該方法對194份臨床豬糞便樣品進行了檢測,結(jié)果顯示PDCoV陽性率為22.1%,明顯高于常規(guī)RT-PCR的11.9%,表明該方法可應(yīng)用于PDCoV的臨床診斷及定量檢測。
[Abstract]:According to the published (PDCoV) N gene sequence of porcine Delta coronavirus (GenBank), specific primers and TaqMan probes were designed and synthesized to construct the standard plasmid. A real-time TaqMan quantitative PCR method for detecting PDCoV was established. The sensitivity, specificity and repeatability of the method were evaluated. The results showed that fluorescent signals were not detected using porcine epidemic diarrhea virus, porcine transmissible gastroenteritis virus, porcine Kub virus, porcine reproductive and respiratory syndrome virus and foot-and-mouth disease virus as templates. The coefficient of variation of circulating threshold Ct was lower than that of 2.66 脳 10 ~ 6 脳 10 ~ (5) and 2.66 脳 10~4copies/L plasmids. The results showed that the method had a good specificity and the coefficient of variation of circulating threshold Ct was lower than that of 2.66 脳 10 ~ (6) and 2.66 脳 10 ~ (6) 10~4copies/L. The slope of the standard curve was -3.461, and the correlation coefficient was 0.998. The results showed that there was a good linear relationship between the threshold value and the concentration of template, and the standard sample was diluted by gradient. The detection of plasmid DNA, with a minimum limit of 2.66 脳 10 ~ (-1) copies/L showed that the method had a good sensitivity. The results showed that the positive rate of PDCoV was 22.1, which was significantly higher than that of routine RT-PCR (11.9%), which indicated that this method could be applied to the clinical diagnosis and quantitative detection of PDCoV.
