香豬睪丸microRNA鑒定及生物學(xué)功能分析
[Abstract]:MicroRNA (miRNA) is a kind of important non-coding small RNA, which regulates the expression of genes at the post-transcriptional level of target genes. It has obvious phase and tissue specificity and plays an important role in the growth and reproduction of animals at various stages of development. As an excellent representative of Guizhou local pig breeds, Xiang pig has early sexual maturity and low litter size. At present, the reproductive regulation mechanism of Xiang pig is not completely clear. In this paper, the microstructures of testis of fragrant pigs at different months of age were studied by histological section technique, and the small RNA c DNA library of testicular tissue of four-month-old Xiang pigs was constructed. HiSeq high-throughput sequencing technique and bioinformatics analysis were used to analyze the microstructures of testis. The expression profiles of miRNA in different developmental stages of Xiang pig were identified. The molecular types and structures of miRNA were analyzed, and the target genes and functions of miRNA were speculated. Six differentially expressed miRNA, studies were selected to study the changes of expression levels in testis and other tissues at different developmental stages. The main results were as follows: 1. The characteristics of testicular development in Xiang pigs were that mature spermatozoa could be detected at the age of 4 months. Using HiSeq high-throughput sequencing technique, the microRNA expression profiles were obtained at the age of 2 months, 3 months old, 7 months and 18 months, respectively. The high quality micro RNA sequence of each expression profile was close to 6 million. 254 known miRNA and 180 candidate mi RNA;3 month-old libraries were identified from 2-month-old Xiangxiang pig testicular RNA library. 251 known mi RNA and 176 candidate miRNA;7 month-old libraries were identified. 227 known miRNA and 120 candidate miRNA;18 month-old libraries were identified for 215 known mi RNA and 100 candidate miRNA.. Six miRNA molecules were randomly selected and confirmed to be expressed in testis tissues of corresponding months by real-time fluorescence quantitative PCR. The results showed that there were significant differences in the expression of 1165 kinds of miRNA in testis library in four periods. 74 miRNA expression levels in 2-month-old libraries were significantly lower than those in 3-month-old libraries and 2-month-old libraries, and the expression levels of miRNA in 2-month-old libraries were significantly higher than those in 7-month-old libraries and 18-month-old libraries. 219 and 224 miRNAs up-regulated the expression of miRNA in three-month-old library compared with 7-month-old library and 18-month-old library, respectively. 268 and 272 miRNAs expression levels were significantly higher than that of 18-month-old library, respectively, and the expression of miRNA in 7-month-old library was significantly higher than that in 18-month-old library. The expression of miRNA in the testis of 21 miRNAs was the highest and the lowest among 18 months of age. The target genes of 10 differentially expressed miRNA were predicted by mirecord software. A total of 1953 target genes were obtained by KEGG Pathway analysis. These target genes may be involved in testicular development and spermatogenesis of Xiang pig. By real-time quantitative RT-PCR detection, 6 mi RNAs were expressed in testicular tissues of different developmental stages, and the general trend was: With the increase of testis from 1 month to 7 months, the expression of Let-7a-5p and miR-10 increased first, then decreased gradually. The expression of miR-200b decreased first and then increased. There was no obvious regularity in the expression of miR-145-5p and miR-199a-3p. In this paper, the differential expression of miRNA in the testis of Xiang pig at different ages was systematically studied, which was helpful to explain the molecular regulation mechanism of testicular development in Xiang pig.
