淫羊藿多糖提取工藝的優(yōu)化及其對肉雞免疫功能的調(diào)節(jié)作用

【摘要】:淫羊藿多糖(Epimedium polysaccharide)是淫羊藿(Epimedium brevicornum Maxim)的主要有效成分之一,具有調(diào)節(jié)免疫功能、抗病毒、抗氧化等多種生物活性和功能。資料表明,淫羊藿多糖是畜禽的一種良好免疫調(diào)節(jié)劑,具有較高的研究價(jià)值和應(yīng)用前景。本文探索研究了淫羊藿多糖的提取工藝、含量測定及其對肉雞生長性能和免疫調(diào)節(jié)作用的影響。1、創(chuàng)建一種淫羊藿多糖含量測定的方法(苯酚-硫酸法),并在此基礎(chǔ)上,通過研究料水比、提取溫度、提取時(shí)間、提取次數(shù)單因素,以及4因素3水平正交優(yōu)化實(shí)驗(yàn),優(yōu)化淫羊藿多糖的提取工藝,并得到最佳的提取條件:料水比為1:25,提取溫度為90℃,提取時(shí)間為2h,提取次數(shù)為3次。同時(shí)研究醇沉濃度,得到70%醇濃度為最佳的醇沉濃度。2、按照最佳提取條件提取淫羊藿多糖,并進(jìn)行醇沉純化,通過應(yīng)用苯酚-硫酸法對淫羊藿多糖含量進(jìn)行測定。測得淫羊藿粗多糖的平均得率為5.81%,淫羊藿粗多糖中多糖的平均含量是42.36%,淫羊藿中多糖平均含量為2.46%。3、將利用最佳條件提取純化的淫羊藿多糖制成不同劑量進(jìn)行動物實(shí)驗(yàn),分為淫羊藿多糖實(shí)驗(yàn)組、免疫抑制組和空白對照組。免疫抑制組應(yīng)用免疫抑制劑環(huán)磷酰胺(CY)進(jìn)行免疫抑制,淫羊藿多糖實(shí)驗(yàn)組分別研究低劑量、中劑量、高劑量淫羊藿多糖對肉雞的生產(chǎn)性能和免疫功能的調(diào)節(jié)作用。結(jié)果表明,不同劑量的淫羊藿多糖對肉雞的平均日增重(ADG)、料肉比(FCR)等生產(chǎn)性能均有不同程度地促進(jìn)作用;不同劑量的淫羊藿多糖對肉雞的免疫器官指數(shù)、免疫球蛋白、細(xì)胞因子均有增強(qiáng)作用;在不同實(shí)驗(yàn)階段,經(jīng)過統(tǒng)計(jì)學(xué)分析,淫羊藿多糖實(shí)驗(yàn)組的免疫指標(biāo)顯著好于免疫抑制組(P0.05),并且,中、高劑量的一些免疫指標(biāo)超過了空白對照組,但尚未達(dá)到差異顯著(P0.05)。4、將利用最佳條件提取純化的淫羊藿多糖制成不同劑量進(jìn)行實(shí)驗(yàn),分為淫羊藿多糖實(shí)驗(yàn)組和免疫抑制組和空白對照組。免疫抑制組應(yīng)用免疫抑制劑環(huán)磷酰胺(CY)進(jìn)行免疫抑制,淫羊藿多糖實(shí)驗(yàn)組分別研究低劑量、中劑量、高劑量淫羊藿多糖對肉雞的腸道形態(tài)和抗氧化功能的影響。結(jié)果表明,不同劑量的淫羊藿多糖均可不同程度地增長肉雞的腸道絨毛長度,降低隱窩深度,增加腸道內(nèi)乳酸桿菌等有益菌的數(shù)量,減少腸道大腸桿菌、沙門氏菌等有害菌的數(shù)量;不同劑量的淫羊藿多糖均可不同程度地增強(qiáng)抗氧化物質(zhì)SOD、GSH-Px、CAT的活性,降低氧化物質(zhì)MDA的活性。并且,在不同實(shí)驗(yàn)階段,淫羊藿多糖不同劑量組和免疫抑制組相比,上述實(shí)驗(yàn)指標(biāo)差異顯著(P0.05),而且,有些劑量的檢測指標(biāo)超過了空白對照組,但尚未達(dá)到差異顯著(P0.05)。

In this paper, the extraction technology of Epimedium polysaccharide, the determination of its content and its effect on growth performance and immunomodulation of broilers were studied. A method for the determination of Epimedium polysaccharide (phenol-sulfuric acid method) was established. The extraction process of Epimedium polysaccharides was optimized by orthogonal experiments of the ratio of material to water, extraction temperature, extraction time, single factor of extraction times and 4 factors and 3 levels.
