蛹蟲草多糖對雞新城疫疫苗免疫效果的影響

【摘要】:為研究蛹蟲草多糖(CMP)對雞新城疫苗免疫效果的影響,將200只14日齡雛雞隨機(jī)分為5組,除空白對照組(BC)外,均接種新城疫疫苗,28日齡第二次接種。第一次接種疫苗的同時,三個CMP40組雞分別肌肉注射低、中、高濃度的多糖,免疫對照組(VC)和BC組注射等量生理鹽水,每天1次,連續(xù)3 d。在第1次接種后的第7(D7)、14(D14)、21(D21)、28(D28)、35(D35)和42(D42)天采血,分別用MTT法和血凝抑制(HI)試驗(yàn)測定外周血淋巴細(xì)胞增殖和血清抗體效價變化,在D14、D21和D28用ELISA測定血清IL-4和IFN-γ濃度。結(jié)果顯示,CMP40L組在所有時間點(diǎn)的A570值和血清抗體效價顯著高于VC組和BC組(P0.05);低劑量的CMP40發(fā)揮佐劑性能的效果最好,同時可促進(jìn)機(jī)體對IFN-γ和IL-4的分泌。
[Abstract]:In order to study the effect of Cordyceps militaris polysaccharide (CMP) on the immune effect of chicken Newcastle vaccine, 200 14-day-old chicks were randomly divided into 5 groups, all of them were inoculated with Newcastle disease vaccine for the second time at 28 days except the blank control group (BC). At the same time, the three CMP40 groups were injected with low, medium and high concentrations of polysaccharides intramuscularly, while the control group, (VC) and BC groups, were injected with the same amount of normal saline once a day for 3 days. Blood samples were collected on the 7th (D7) 14 (D14) 21 (D21) 28 (D28) -35 (D35) and 42 (D42) days after the first inoculation. The proliferation of peripheral blood lymphocytes and serum antibody titers were measured by MTT assay and hemagglutination inhibition (HI) test. Serum IL-4 and IFN- 緯 concentrations were measured by ELISA at D14D21 and D28, respectively. The results showed that the A570 value and serum antibody titer in CMP 40L group were significantly higher than those in VC group and BC group at all time points (P0.05). Low dose CMP40 had the best effect on adjuvant performance and could promote the secretion of IFN- 緯 and IL-4.
