嗜酸乳桿菌發(fā)酵飼料及其對母豬健康和繁殖性能的影響

【摘要】:本研究對一株豬源嗜酸乳桿菌的體外生物學特性進行了分析,并優(yōu)化篩選出其最佳凍干保護劑配方。使用凍干制劑對滅菌豆粕、普通豆粕和哺乳母豬配合飼料三種基質進行固態(tài)發(fā)酵,優(yōu)化得到了三種基質的最佳發(fā)酵工藝參數(shù),檢測和分析了發(fā)酵前后營養(yǎng)成分變化。并開展了母豬飼喂試驗。1.嗜酸乳桿菌最適接種量為4%;培養(yǎng)60h時產(chǎn)乳酸量最高,為66.85 mmol/L;在MRS液體培養(yǎng)基pH=2.0時,培養(yǎng)4h存活率達70.14%;在0.2%、0.3%豬膽鹽條件下保溫2h時活菌數(shù)分別為6.11和4.53 log(CFU/m L);對有害的大腸桿菌、沙門氏菌和金黃色葡萄球菌的抑菌圈直徑均在19 mm以上;菌懸液濃度為109CFU/m L時,該菌對豬腸上皮細胞的黏附指數(shù)最高為1060CFU/100cell。2.該菌株的凍干保護劑最佳配方為:脫脂乳16.216%、乳糖4.375%、L-谷氨酸鈉0.123%、L-半胱氨酸0.143%、糊精9.469%,凍干保護率可達93.91%。3.滅菌豆粕最佳發(fā)酵工藝參數(shù)為:發(fā)酵溫度37℃,料水比1:1.2,接種量0.8%,時間36h,嗜酸乳桿菌活菌數(shù)為9.10 lg(CFU/g);普通豆粕最佳發(fā)酵工藝參數(shù)為:發(fā)酵溫度37℃,料水比1:1.2,接種量1.0%,時間48h,活菌數(shù)為9.02 lg(CFU/g);哺乳母豬配合飼料最佳發(fā)酵工藝參數(shù)為:發(fā)酵溫度37℃,料水比1:0.8,接種量0.4%,時間36h,活菌數(shù)為9.48 lg(CFU/g)。發(fā)酵后,滅菌豆粕、普通豆粕和配合飼料中的活菌數(shù)均由發(fā)酵前的106 CFU/g達到109CFU/g,揮發(fā)性鹽基氮含量均小于50 mg/100g,其中粗蛋白含量分別提高2.16%、1.80%和13.10%,酸溶蛋白含量分別提高10.36%、11.73%和20.62%,鈣含量分別提高3.13%、3.13%和3.23%,總磷含量分別提高2.78%、2.82%和4.92%。4.母豬試驗表明,與對照組比較,母豬日糧中添加固態(tài)發(fā)酵飼料,使仔豬出生成活率、斷奶成活率和仔豬平均日增重分別提高了2.60%、9.20%和9.10%,仔豬腹瀉率降低53.24%,哺乳母豬平均日采食量提高了13.79%;哺乳母豬血清中Ig G含量提高7.58%;初乳中蛋白質、總固形物、Ig A和Ig M的含量分別提高18.08%、1.30%、4.06%和5.61%;鮮糞中乳酸菌數(shù)量由7.59 lg(CFU/g)提高到9.04 lg(CFU/g)。
[Abstract]:The biological characteristics of a porcine Lactobacillus acidophilus strain in vitro were analyzed and the optimum lyophilization protectant formula was optimized. The solid state fermentation of sterilized soybean meal, common soybean meal and lactation sows was carried out by using freeze-dried preparation. The optimum fermentation parameters of the three substrates were optimized, and the changes of nutritional components before and after fermentation were detected and analyzed. The experiment of sows feeding was also carried out. The optimum inoculation amount of Lactobacillus acidophilus was 4, the lactic acid production was 66.85 mmol/L; at 60 h and the survival rate reached 70.14 when cultured on MRS liquid medium pH=2.0, and the number of live bacteria was 6.11 log (CFU/m L);) and 4.53 log (CFU/m L); was harmful to harmful Escherichia coli) when incubated with 0.3% bile salt for 2 hours.
