飼喂發(fā)酵棉粕肉牛牛糞中細(xì)菌數(shù)量及多樣性研究

【摘要】:【目的】研究飼喂發(fā)酵棉粕對(duì)安格斯肉牛牛糞中細(xì)菌數(shù)量及多樣性的影響。【方法】采用平板計(jì)數(shù)法計(jì)算細(xì)菌的數(shù)量;采用PCR技術(shù)對(duì)提出的DNA進(jìn)行細(xì)菌16Sr DNA基因V6-V8區(qū)擴(kuò)增,將PCR產(chǎn)物進(jìn)行變性梯度凝膠電泳(DGGE)。得到指紋圖譜特異性條帶切膠回收并進(jìn)行克隆測(cè)序,通過BLAST鑒定細(xì)菌菌種!窘Y(jié)果】(1)試驗(yàn)前期,C組和T組總細(xì)菌、大腸桿菌和乳酸菌差異顯著(P0.05);C組和T組金黃色葡萄球菌差異不顯著(P0.05)。試驗(yàn)后期,C組和T組總細(xì)菌差異不顯著(P0.05);C和T組大腸桿菌、金黃色葡萄球菌和乳酸菌差異顯著(P0.05)。(2)牛糞中存在的菌種有沙門氏菌(Salmonella)、瘤胃球菌(Ruminococcus)、乳酸菌(Lactobacillus)、毛螺旋菌(Lachnospiraceae)、志賀菌(Shigella)、梭菌(Clostridium)、大腸桿菌(Escherichia coli)、腸桿菌(Enterobacter)和腸球菌(Enterococcus),而其中乳酸菌、大腸桿菌可能是牛糞中的優(yōu)勢(shì)菌群!窘Y(jié)論】試驗(yàn)前期和后期總細(xì)菌、大腸桿菌、金黃色葡萄菌的數(shù)量有明顯變化,牛糞中存在多種細(xì)菌表現(xiàn)了牛糞中微生物菌群的多樣性。
[Abstract]:[objective] to study the effect of fermented cottonseed meal on the number and diversity of bacteria in Angus cattle dung. [methods] the number of bacteria was calculated by plate counting method, and the V6-V8 region of bacterial 16Sr DNA gene was amplified by PCR technique. Denaturing gradient gel electrophoresis (DGGE).) of PCR products The specific band cut gum of fingerprint was obtained and cloned and sequenced. The bacterial species were identified by BLAST. [results] (1) the total bacteria of C group and T group in the early stage of the test. There was no significant difference in Escherichia coli and lactic acid bacteria (P0.05) between group C and group T (P0.05). There was no significant difference in total bacteria between group C and group T (P0.05). There were significant differences between Staphylococcus aureus and lactobacillus (P0.05). (2) among which salmonella (Salmonella), (Salmonella), (Ruminococcus), lactic acid bacteria, (Lactobacillus), (Lachnospiraceae), (Shigella), (Shigella), (Clostridium), Escherichia coli, (Escherichia coli), Enterobacter (Enterobacter), and enterococcal (Enterococcus), among them, lactic acid bacteria. [conclusion] the number of total bacteria, Escherichia coli and grapevine aureus in the early and late stages of the experiment were obviously changed, and the variety of bacteria in cattle manure showed the diversity of microflora in cow manure.
