體外消化法優(yōu)化生長(zhǎng)豬玉米-豆粕-DDGS飼糧和小麥-豆粕飼糧非淀粉多糖酶譜的研究

【摘要】:旨在探討使用體外法模擬豬飼糧胃腸道消化,優(yōu)化生長(zhǎng)豬玉米-豆粕-DDGS飼糧和小麥-豆粕飼糧的非淀粉多糖(NSP)酶譜。試驗(yàn)首先采用單因素完全隨機(jī)設(shè)計(jì),在玉米-豆粕-DDGS飼糧和小麥-豆粕飼糧中分別添加不同水平的單一NSP酶:纖維素酶、木聚糖酶、β-甘露聚糖酶、α-半乳糖苷酶、β-葡聚糖酶和果膠酶,采用基于生長(zhǎng)豬生理消化的模擬胃液-小腸液體外法對(duì)飼糧體外干物質(zhì)消化率(in vitro dry matter digestibility,IVDMD)進(jìn)行測(cè)定,分析各NSP酶對(duì)飼糧IVDMD的作用效果。然后采用二次回歸通用旋轉(zhuǎn)設(shè)計(jì),分別建立兩種飼糧IVDMD與6種NSP酶的回歸關(guān)系,篩選兩種飼糧中6種NSP酶的最佳組合,再對(duì)優(yōu)化的酶譜組合進(jìn)行體外驗(yàn)證。結(jié)果顯示:1)生長(zhǎng)豬玉米-豆粕-DDGS飼糧和小麥-豆粕飼糧的IVDMD與6種NSP酶的添加水平之間均存在二次曲線關(guān)系;2)在本試驗(yàn)條件下,玉米-豆粕-DDGS飼糧的最佳酶譜:纖維素酶427.1U·kg~(-1)、木聚糖酶7 057.5U·kg~(-1)、β-甘露聚糖酶3 587.8U·kg~(-1)、α-半乳糖苷酶202.1U·kg~(-1)、β-葡聚糖酶1 543.3U·kg~(-1)和果膠酶72.7U·kg~(-1)。小麥-豆粕飼糧的最佳酶譜:纖維素酶1 117.9 U·kg~(-1)、木聚糖酶35 087.7 U·kg~(-1)、β-甘露聚糖酶1 917.1U·kg~(-1)、α-半乳糖苷酶305.0U·kg~(-1)、β-葡聚糖酶806.7U·kg~(-1)和果膠酶133.7U·kg~(-1);3)優(yōu)化后的NSP酶譜組合使玉米-豆粕-DDGS飼糧的IVDMD提升了3.89%;使小麥-豆粕飼糧的IVDMD提升了3.48%。結(jié)果表明,體外法優(yōu)化后的NSP酶譜組合和配伍能更大程度地提高生長(zhǎng)豬玉米-豆粕-DDGS飼糧和小麥-豆粕飼糧的IVDMD,基于生長(zhǎng)豬生理消化的模擬胃液-小腸液體外消化法可快速篩選出生長(zhǎng)豬飼糧合理的NSP酶譜。
[Abstract]:The aim of this study was to optimize the (NSP) zymogram of non-starch polysaccharides (NPS) in growing porcine corn-soybean dregs and wheat-soybean meal diets by simulating the digestion of gastrointestinal tract in pig diets in vitro. In the experiment, a single NSP enzyme, cellulase, was added to corn-soybean meal diet and wheat-soybean meal diet, respectively. Xylanase, 尾 -mannanase, 偽 -galactosidase, 尾 -glucanase and Pectinase were used to determine the dry matter digestibility (in vitro dry matter digestibility,IVDMD) of diet in vitro by simulated gastric liquid-small intestinal fluid method based on physiological digestion of growing pigs. The effects of NSP enzymes on dietary IVDMD were analyzed. Then the quadratic regression general rotation design was used to establish the regression relationship between two dietary IVDMD and six NSP enzymes, and the optimal combination of six NSP enzymes in the two diets was selected, and the optimized zymogram combination was verified in vitro. The results showed that there was a conic relationship between the levels of IVDMD and six NSP enzymes in the corn-soybean dregs and wheat-soybean meal diets of growing pigs. The optimum zymogram of corn-soybean dregs diet: cellulase 427.1 U kg~ (-1), xylanase 7 057.5 U kg~ (-1), 尾 -mannanase 3 587.8 U kg~ (-1), 偽 -galactosidase 202.1 U kg~ (-1), 尾 -glucanase 1 543.3 U kg~ (-1) and Pectinase 72.7 U kg~ (-1). Optimum zymogram of wheat-soybean meal diet: cellulase 1 117.9 U kg~ (-1), xylanase 35 087.7 U kg~ (-1), 尾 -mannanase 1 917.1 U kg~ (-1), 偽 -galactosidase 305.0 U kg~ (-1), 尾 -glucanase 806.7 U kg~ (-1) and Pectinase 133.7 U kg~ (-1) optimized NSP zymogram combination of corn-soybean dregs-DDGS diet IVDMD The IVDMD of wheat-soybean meal diet was increased by 3. 48%. The results show that The combination and compatibility of NSP zymogram optimized by in vitro method can greatly improve the IVDMD, of growing pig corn-soybean dregs and wheat-soybean meal diets based on the simulated gastric juice and intestinal fluid extradigestion method based on physiological digestion of growing pigs. The reasonable NSP zymogram of growing pig diet was screened out.
