【摘要】：為研究交感神經(jīng)對(duì)妊娠早期子宮內(nèi)DKK-1和Wnt-1的調(diào)控作用,選取180~200日齡健康的性成熟Wistar雌性大鼠,隨機(jī)分為對(duì)照組和處理組,處理組采用靜脈注射6-OHDA損毀交感神經(jīng),用免疫組化法和qRT-PCR法,觀察和分析交感神經(jīng)對(duì)妊娠早期大鼠子宮內(nèi)膜的DKK-1和Wnt-1蛋白及mRNA相對(duì)表達(dá)量的影響。結(jié)果顯示:6-OHDA損毀交感神經(jīng)后,對(duì)卵巢排卵數(shù)無顯著性影響,但子宮囊胚數(shù)量顯著下降、囊胚發(fā)育延遲,導(dǎo)致子宮腔上皮和腺上皮細(xì)胞自分泌DKK-1和Wnt-1紊亂,呈現(xiàn)與正常狀態(tài)相反的變化趨勢(shì)。結(jié)果表明:在著床窗口期,交感神經(jīng)通過影響DKK-1對(duì)Wnt-1信號(hào)分子的調(diào)控作用而影響囊胚著床、發(fā)育。
[Abstract]:In order to study the effect of sympathetic nerve on DKK-1 and Wnt-1 in the uterus of early pregnancy, female rats with sexual maturity of 180 ~ 200 days old were randomly divided into control group and treatment group. The treatment group was treated with intravenous 6-OHDA to damage the sympathetic nerve. The effects of sympathetic nerve on the expression of DKK-1, Wnt-1 protein and mRNA in endometrium of early pregnancy rats were observed and analyzed by immunohistochemical method and qRT-PCR method. The results showed that the number of ovarian ovulation was not significantly affected by the lesion of the sympathetic nerve, but the number of uterine blastocysts decreased significantly, and the development of blastocysts was delayed, which resulted in the disturbance of DKK-1 and Wnt-1 in uterine cavity epithelium and glandular epithelium. Presents a trend of change contrary to the normal state. The results showed that the sympathetic nerve affected blastocyst implantation and development by affecting the regulation of Wnt-1 signaling molecules by DKK-1 during implantation window.
