阿魏根粗多糖對雞脾臟和外周血淋巴細胞體外增殖作用的影響

【摘要】:為研究新疆阿魏多糖(FSP)對雞外周血T淋巴細胞和脾臟B淋巴細胞增殖作用的影響。本研究用水提醇沉法得到阿魏總多糖(FSPt)和3種分級多糖(FSP30、FSP50、FSP70),并通過苯酚—硫酸法和紅外光譜對FSP進行檢測。通過MTT法測定各級FSP的安全濃度及其對雞外周血T淋巴細胞及脾臟B淋巴細胞的增殖能力。結果顯示,提取的FSP均具有多糖的特征峰,為多糖類化合物。外周血T淋巴細胞增殖試驗表明,除FSP50外,所有多糖在一定濃度下均能單獨和協(xié)同PHA刺激T淋巴細胞增殖,其中FSPt的增殖作用較強。脾臟B淋巴細胞增殖試驗表明,除FSP50外,所有多糖在一定濃度下均能單獨和協(xié)同LPS刺激B淋巴細胞增殖,其中FSP70和FSPt增殖作用較強。綜上所述,各級FSP中,FSPt對脾臟B淋巴細胞和外周血T淋巴細胞的增殖作用最強,可以作為進一步研究的材料。
[Abstract]:To study the effect of Xinjiang Ferula polysaccharide (FSP) on the proliferation of T lymphocytes and B lymphocytes in chicken peripheral blood. In this study, the total polysaccharide of ferulic (FSPt) and three kinds of graded polysaccharides (FSP30,FSP50,FSP70) were obtained by water extraction and alcohol precipitation, and FSP was determined by phenol-sulfuric acid method and infrared spectrum. The safe concentration of FSP at all levels and its proliferation ability to T lymphocytes and B lymphocytes in peripheral blood of chicken were determined by MTT method. The results showed that the extracted FSP had the characteristic peak of polysaccharides and was a polysaccharide compound. The T lymphocyte proliferation test in peripheral blood showed that all polysaccharides except FSP50 could stimulate T lymphocyte proliferation alone and in coordination with PHA, and FSPt had stronger proliferative effect. Splenic B lymphocyte proliferation test showed that all polysaccharides except FSP50 could stimulate B lymphocyte proliferation alone and in coordination with LPS. FSP70 and FSPt had stronger proliferative effects. In conclusion, the proliferation of spleen B lymphocytes and peripheral blood T lymphocytes by FSPt was the strongest in all levels of FSP, and could be used as a material for further study.
