胍基乙酸的致突變性研究

【摘要】:本文選用Ames試驗(yàn)、小鼠骨髓細(xì)胞微核試驗(yàn)和精子畸形試驗(yàn)三種短期測試組合探究胍基乙酸是否具有致突變作用。結(jié)果發(fā)現(xiàn):Ames試驗(yàn)結(jié)果顯示,胍基乙酸在8~5000μg/皿,有或無代謝活化系統(tǒng)(S9)時(shí),對鼠傷寒沙門氏菌TA_(97)、TA_(98)、TA_(100)、TA_(102)四種試驗(yàn)株均沒有誘變性;骨髓細(xì)胞微核試驗(yàn)結(jié)果顯示,胍基乙酸在1250~5000 mg/kg體重劑量內(nèi)經(jīng)口染毒后,各劑量組小鼠骨髓細(xì)胞中嗜多染紅細(xì)胞(PCE)微核率與陰性對照組比較均無顯著性差異(P0.05);小鼠精子畸形試驗(yàn)結(jié)果顯示,胍基乙酸在1250~5000 mg/kg體重劑量內(nèi)經(jīng)口染毒后,各劑量組小鼠精子畸形率與陰性對照組比較均無顯著性差異(P0.05)。結(jié)果表明,三個(gè)致突變試驗(yàn)結(jié)果均為陰性,胍基乙酸為非致突變物質(zhì)。
[Abstract]:In this paper, Ames test, mouse bone marrow micronucleus test and sperm malformation test were used to study the mutagenicity of guanidoacetic acid. The results showed that guanidinoacetic acid had no mutagenicity on TA_ _ (97) Tap _ (100) TA102.The results of micronucleus test of bone marrow cells showed that there was no mutagenicity in the presence or absence of metabolic activation system (S9) for guanidinoacetic acid (GAA) in the presence or absence of metabolic activation system (S9), and the results of micronucleus test in bone marrow cells showed that there was no mutagenicity in the four test strains of Salmonella typhimurium, There was no significant difference in micronucleus rate of polychromatic erythrocyte (PCE) in bone marrow cells between the two groups after oral administration of guanidine acetic acid at the dose of 1250,5000 mg/kg (P0.05), and the results of sperm malformation test showed that the micronucleus rate of polychromatic erythrocytes in the bone marrow cells of the mice was higher than that of the control group (P0.05). There was no significant difference in sperm deformity rate between each dose group and negative control group after oral administration of guanidine acetic acid at 1250 mg/kg body weight (P0.05). The results showed that all the three mutagenicity tests were negative and guanidine acetic acid was a non-mutagenic substance.
