【摘要】：以10 mg/kg體重的劑量研究豬內(nèi)服鹽酸多西環(huán)素飲水劑的藥動學(xué)特征,采用超高效液相色譜-串聯(lián)質(zhì)譜法,測定豬內(nèi)服與靜脈給予多西環(huán)素的血漿濃度,Winnolin軟件進行分析。豬單次內(nèi)服多西環(huán)素飲水劑型水溶液后,MRT_(last)為(12.82±3.34)h,T1/2為(11.65±4.80)h,T_(max)為(4.13±1.89)h,Cmax為(0.87±0.48)μg/m L,AUC_(last)為(11.07±4.19)μg·h·mL~(-1)。靜脈注射10 mg/kg體重多西環(huán)素水溶液后,AUClast為(33.34±8.03)μg·h·mL~(-1),T1/2為(5.68±1.32)h,MRT_(last)為(7.98±0.56)h。結(jié)果表明多西環(huán)素飲水劑水溶液的絕對生物利用度為33.2%。豬內(nèi)服多西環(huán)素飲水劑后,具有體內(nèi)吸收速度較快,達峰時間短,消除較慢,血中平均滯留時間較長等特點。
[Abstract]:The pharmacokinetic characteristics of doxycycline hydrochloride drinking water in pigs were studied at the dose of 10 mg/kg body weight. The plasma concentration of doxycycline hydrochloride in pigs was determined by ultra-high performance liquid chromatography-tandem mass spectrometry (HPLC-MS). The software Winnolin was used to analyze the pharmacokinetics of doxycycline hydrochloride. The MRT _ (last) was (12.82 鹵3.34) h ~ (-1) and (11.65 鹵4.80) h ~ (-1) (max) was (0.87 鹵0.48) 渭 g / m ~ (-1) 渭 g / L AUC _ (last), (11.07 鹵4.19) 渭 g / h mL~ (-1). AUC last was (33.34 鹵8.03) 渭 g / h mL~ (-1) T _ 1 / 2 after intravenous injection of doxycycline solution for 10 mg/kg (5.68 鹵1.32) h MRT _ (last) was (7.98 鹵0.56) h. The results showed that the absolute bioavailability of doxycycline water solution was 33.2. After pigs were given doxycycline drinking water, the absorption rate was faster, the peak time was short, the elimination was slower, and the average retention time in blood was longer.
