灌注酵母培養(yǎng)物對(duì)綿羊血液生化指標(biāo)、免疫及抗氧化功能的影響

【摘要】:本研究目的是對(duì)不同部位灌注不同劑量酵母培養(yǎng)物對(duì)綿羊血液生化指標(biāo)、免疫及抗氧化功能影響進(jìn)行比較研究,首先通過(guò)體外模擬瘤胃法篩選酵母培養(yǎng)物最適添加量;然后進(jìn)行綿羊體內(nèi)灌注試驗(yàn),研究酵母培養(yǎng)物的作用效果和不同部位的效果差異;最后對(duì)灌注試驗(yàn)結(jié)果進(jìn)行綜合比較。選用9只體況良好、體重相近,裝有瘤胃、十二指腸和回腸瘺管的小尾寒羊,隨機(jī)分3組,每組3個(gè)重復(fù),每個(gè)重復(fù)1只羊。I組為對(duì)照組,II組為體外最適宜添加量組,III組為體外第二適宜添加量組,在基礎(chǔ)日糧條件下瘤胃、十二指腸、回腸分期灌注占日糧干物質(zhì)0.0%、1.5%和2.0%的酵母培養(yǎng)物。其中瘤胃灌注試驗(yàn)預(yù)飼期14d,正式期21d,十二指腸和回腸灌注試驗(yàn)預(yù)飼期14d,正式期14d。論文共分為5個(gè)部分:1:體外模擬瘤胃法篩選酵母培養(yǎng)物的最適添加量。以試驗(yàn)日糧作為培養(yǎng)底物,并分別添加占培養(yǎng)底物干物質(zhì)的0.0%、1.0%、1.5%、2.0%的酵母培養(yǎng)物。結(jié)果表明,1.5%添加量效果最好,2.0%添加量次之。其中各組pH只有12h出現(xiàn)顯著差異,隨著酵母培養(yǎng)物添加量的增加有下降趨勢(shì),2.0%組最低(P0.05);各組NH3-N在不同時(shí)間影響程度不同:6h時(shí)1.5%和2.0%組較低,12h時(shí)1.0%和1.5%較低,24h時(shí)對(duì)照組和1.0%較低;從整體趨勢(shì)看,12h和24h的VFA變化趨勢(shì)相同,與6h存在差異。2:瘤胃灌注酵母培養(yǎng)物結(jié)果表明:酵母培養(yǎng)物能夠顯著提高綿羊血清TP、GLU水平(P0.05),顯著降低血清TG和UREA含量(P0.05),ALP有升高的趨勢(shì);能夠不同程度提高血清中T-AOC,SOD和GSH-Px水平,并減少M(fèi)DA生成量;第7d和21d極顯著提高了綿羊顯著提高了血清IgG含量(P0.01),2.0%組IgA含量在灌注第21d時(shí)顯著高于對(duì)照組(P0.05);顯著提高了IL-2、IL-6含量(P0.05),IFN-γ含量達(dá)到極顯著水平(P0.01)。3:十二指腸灌注酵母培養(yǎng)物結(jié)果表明:通過(guò)十二指腸灌注酵母培養(yǎng)物,顯著提高綿羊血清GLU含量(P0.05)。提高了綿羊血清T-AOC和T-SOD含量,降低MDA含量(P0.05);能夠提高IgA、IgG和IL-6的含量,顯著提高IL-2、IFN-γ含量(P0.05)。4:回腸灌注酵母培養(yǎng)物結(jié)果表明:回灌注酵母培養(yǎng)物有提高ALB和GLU含量的趨勢(shì);未對(duì)綿羊抗氧化指標(biāo)產(chǎn)生顯著的影響;隨著灌注天數(shù)增加有提高IL-6含量的趨勢(shì)。5:綜合比較結(jié)果表明:本試驗(yàn)酵母培養(yǎng)物在綿羊上應(yīng)用有實(shí)際效果,能夠改善血液指標(biāo),提高抗氧化能力,提高免疫能力;不同灌注劑量之間并且未出現(xiàn)明顯的效果差異,本試驗(yàn)條件下瘤胃灌注酵母培養(yǎng)物即可達(dá)到最佳效果,不需要就行過(guò)瘤胃保護(hù)處理。
[Abstract]:The purpose of this study was to compare the effects of different doses of yeast culture on blood biochemical indexes, immunity and antioxidant function of sheep. Then the effects of yeast culture and different parts of the yeast culture were studied. Finally, the results of the perfusion test were compared synthetically. Nine small tail Han sheep with rumen, duodenum and ileal fistula were randomly divided into 3 groups. Group I was the control group, group II was the best dosage group in vitro, group III was the second optimal dosage group in vitro. Under the basic diet condition, the rumen, duodenum and ileum were infused with 0.01.5% and 2.0% of the dry matter in the diet by stages. Among them, rumen perfusion test was prefed for 14 days, formal for 21 days, duodenum and ileum for 14 days, and formal for 14 days. The thesis was divided into five parts: 1: the optimum addition of yeast culture was screened by rumen mimic method in vitro. The experimental diet was used as the culture substrate, and the yeast culture consisted of 0.010% of the dry matter of the culture substrate and 1.5% and 2.0% of the dry matter of the culture substrate respectively. The results showed that 1.5% addition had the best effect of 2.0% addition. Among them, the pH value of each group was significantly different only at 12 h, and the lowest was in 2.0% group with the increase of yeast culture addition (P0.05), and the NH3-N was lower in 1.5% and 2.0% groups than in the control group at 12 h and 1.5% at 12 h and 1.0% at 24 h, respectively, at different time and at 6 h after addition of yeast (P0.05), and the control group was 1.0% and 1.0% lower than the control group at 12 h and 1.5%, respectively. According to the whole trend, the change trend of VFA in 12 h and 24 h was the same, and there was a difference between them at 6 h. The results showed that yeast culture could significantly increase serum TPGLU level (P0.05) and decrease serum TG and UREA content (P0.05). It can increase the levels of T-AOC SOD and GSH-Px in serum and reduce the amount of MDA in different degree. On the 7th and 21st days, the serum IgG content was significantly increased in sheep (P0.01). The IgA content in 2.0% group was significantly higher than that in the control group on the 21st day after perfusion (P0.05), and the IL-2IL-6 content (P0.05) was significantly increased in sheep (P0.01). 3: duodenal perfusion yeast culture. The results showed that yeast culture was infused through duodenum. The serum GLU content of sheep was significantly increased (P0.05). The contents of T-AOC and T-SOD in serum of sheep were increased, the contents of MDA were decreased (P0.05), and the contents of IgA T-AOC and IL-6 were increased significantly (P0.05). 4. The results showed that the contents of ALB and GLU were increased in the culture of ileal perfusion yeast. There was no significant effect on the antioxidant index of sheep, and with the increase of perfusion days, there was a tendency to increase the content of IL-6. 5. The comprehensive comparison showed that the yeast culture in this experiment had practical effect on sheep, and could improve the blood index. The effect of yeast culture on rumen perfusion could reach the best effect under the condition of this experiment, without the need of rumen protection treatment, the antioxidation ability was improved and the immune ability was improved, and there was no obvious difference in the effect between the different perfusion doses and there was no obvious difference in the effect.
